The project integrates proteomics, molecular virology, genomics, and bioinformatics to address Cotton Leafroll Dwarf Virus (CLRDV) and Cotton Virus A (COTV-A), which threaten U.S. cotton production. Deliverables include identification and characterization of aphid proteins regulating CLRDV transmission; determination of biological factors controlling COTV-A transmission through aphids, seeds, and endogenous viral elements in cotton genomes; and characterization of population dynamics, genetic diversity, and geographic distribution of CLRDV, COTV-A, and additional viral pathogens in cotton and weed reservoirs across U.S. cotton production regions. The project employs affinity purification-mass spectrometry, reverse-genetics systems, infectious clones, nanobody-based diagnostics, aphid transmission assays, high-throughput sequencing, and virome analyses to discover and evaluate viral populations. Work is performed in Blacksburg, Virginia, with a period of performance from March 1, 2026, through February 28, 2027.
