Description
- Purpose
- The primary purpose of this agreement is to better understand the scope and pathogenesis of the viroids in potato and another solanaceous trap crop, litchi tomato, improve on existing technologies for detection of pospiviroids and their genetic variants, transfer the technologies and help build an infrastructure to improve the capabilities and response of regulatory programs and growers with timely information on best management and eradication practices. The overall goal of this project is to ensure protection of the potato industry in idaho and across the country from an inadvertent introduction of viroids into production areas, and to develop tools for their eradication if an outbreak occurs. 2.
- Activities
- A. To determine the pathogenesis and time-course of the tomato chlorotic dwarf viroid (tcdvd) (and potato spindle tuber viroid (pstvd)) infection in potato and in LT, and to reveal tcdvd (and pstvd) symptomatology in additional solanaceous hosts, such as tobacco, tomato, and nicotiana benthamiana. Optimize tcdvd (and pstvd) detection. B. To determine tcdvd (and pstvd) transmissibility, both mechanical and through plant debris in potato and other hosts, determine tcdvd (and pstvd) longevity in soil. C. To conduct viroid surveillance of the entire greenhouse complex on-campus (ensuring there are no local sources of infection). 3. Deliverables and expected outcomes: several practical questions will be answered, which are proposed as the objectives of this project. The overall goal is to ensure protection of the potato industry in idaho and across the country from an inadvertent introduction of viroids into production areas, and to develop tools for their eradication if an outbreak occurs. The objectives will address the pathogenesis of tcdvd in potato cultivars grown in idaho, the efficiency of mechanical transmission of tcdvd, and its stability in plant residue and in soil. The potato viroid, pstvd will be used as control side-by-side with tcdvd, to compare the phenotypes of infected plants and determine the degree of specificity of different RT-PCR tests. 4. Intended beneficiary(IES): beneficiaries include the potato industry in general as well as individual growers 5. Subrecipient activities, if known or specified at the time of award: $5,882 will be sent to USDA ARS in new york to maintain and propagate the 15 wild potato lines in their greenhouse, and will provide them as tissue culture plantlets to the university of idaho laboratory for viroid screening.
