About this award

The National Aeronautics and Space Administration (NASA) has awarded a $298,974.04 grant to the University of Wisconsin - Madison, operating as Research and Sponsored Programs under the University of Wisconsin System, to conduct research on the effects of altered gravity on nuclear morphology and cytoskeletal organization in fibroblasts during parabolic flight. The research will involve culturing cells in specialized hardware to control temperature and humidity, fixing the cells at different time points, and using advanced imaging and analysis techniques to characterize the mechanical forces acting on the nucleus and identify differentially expressed genes in response to altered gravity. This grant-funded research project does not have a set-aside designation and is focused on furthering scientific understanding of how cells respond to changes in the gravity environment, which is relevant to NASA's space exploration and research mission.

Description

We propose to identify the effects of altered gravity on nuclear morphology and cytoskeletal organization in fibroblasts flown in parabolic flight. Cells flown will be cultured in a specially designed hardware to control for temperature and humidity, and the cells will be fixed on flight at different time points. High resolution imaging of cell shape and nuclear shape will be carried out using a laser scanning confocal microscope. Image analysis software will be used for three- dimensional shape reconstruction and quantification of shape parameters. We will characterize the mechanical forces acting on the nucleus using a fret probe biosensor to investigate the effect of altered gravity on the nuclear tension. Finally, we will identify the genes that are differentially expressed in linc disrupted cells to understand the mechanotransduction pathway in sensing altered gravity.
