Statement of Work (SOW)

Project: Ribonucleic Acid Sequencing (RNAseq) Service

GENERAL INFORMATION

1.0 Scope of Work:

The service provider will perform RNAseq analysis of samples provided by Plant Genetics Research Unit located at the University of Missouri, Columbia, Missouri.

The goal of the ARS project is to identify specific genes that controls the levels of protein content among different soybean cultivars. The specific objective is to monitor changes in gene expression in soybean seeds that have different concentrations of seed proteins.

2.0 Background:

Soybeans exhibit a range in seed protein and oil concentrations. We have limited knowledge on various factors that regulate the seed protein content. We are performing basic research to identify genes and gene functions related to seed composition, particularly high protein. Transcriptome profiling of soybean seeds with different concentration of seed protein levels should facilitate the identification of specific genes that regulate protein concentration. For this reason, we are interested in performing RNAseq analysis. Currently, several companies are providing this service and we would like to utilize one of these companies to perform RNAseq of soybean samples.

CONTRACTOR REQUIREMENTS

2.0 Technical Requirements/Tasks:

54 total RNA samples will be shipped to the service provider on dry ice.

The service provider will perform these specific tasks:

1. Generate standard libraries appropriate for RNA-seq using the 54 provided total

RNA samples.

2. Ensure that resulting libraries meet standard sequencing center QC standards for size, quality, etc.

3. Alignment, classification, & functional annotation of all mapped reads
