Real-Time PCR - Urbana

Statement of Work (SOW)

Project: Real-Time PCR System – Urbana SMGPGRU

GENERAL INFORMATION

1.0 Scope of Work: Acquire a real-time PCR system to increase data analysis throughput and efficiency of the staff studying the molecular biology of plant pathogens and host pathogen interactions at the USDA-ARS SMGPGRU in Urbana, IL.

2.0 Background: The Urbana USDA-ARS SMGPGRU and the USDA Soybean

Germplasm Collection currently house approximately 22,000 accessions of soybean which constitutes the nationally held genetic diversity of the soybean crop. Plant pathologists and geneticists within the unit continually screen the germplasm accessions for pathogen and pest resistance. Once resistance is identified, the underlying genetic nature of the resistance is of utmost importance to investigate and explain. Results from the analysis of real-time PCR systems are critical to the rapid and efficient development of genetic tools to improve the rate of moving disease resistance traits into new genetic backgrounds while also determining the allelic and genic areas of interest.

CONTRACTOR REQUIREMENTS

3.0 Technical Requirements/Tasks: The real-time PCR system will be used for many molecular biology analyses including gene expression analysis, microRNA analysis, single nucleotide polymorphism (SNP) analysis, copy number variation analysis, and allelic discrimination. Below are the technical specifications required.

• Set up for 384 well plates, with the ability to quickly change to 96-well if needed in the future without using tools or additional calibration

• Thermal block which provides consistent temperature distribution, rapid heat equalization, and the ability to run 384 well experiments in less than 40 minutes.

• Broad spectrum, high intensity LED lamp o Ability to acquire raw data o Capture data all in one run, without performing scans o Ability to use a wide range of fluorescent dyes and detection formats

• Built-in data analysis software o Built-in analysis templates o Ability to perform basic or more advanced analysis for gene expression and genotyping functions o Able to visual the plate when editing sample information

• SNP analysis via software should be possible by endpoint or melting curve methods

• Stand-alone, dedicated PC for machine operation and analysis.

o Does not include a built in screen display.

• Has the ability to have barcode plate scanning and plate loading added if the need arises in the future.

4.0 Government Furnished: Not applicable.

5.0 Deliverables / Schedule: The real-time PCR system and any additional parts should be delivered before October 2024.

6.0 Travel: Not applicable.

7.0 Contractor’s Key Personnel: Not applicable.

8.0 Security Requirements: All moving mechanical parts should be isolated from user interaction.

9.0 Data Rights: Not applicable.
