Statement of Work (SOW) Project: Imaging system for chemiluminescent, RGB fluorescent, colorimetric detection of membrane blots, gels, plates, samples
GENERAL INFORMATION
1.0 Scope of Work: The USDA-ARS St. Paul, Minnesota location is requesting quotes for an imaging system for visualization of chemiluminescent, fluorescent, colorimetric membrane blots, gels, plates, and other samples.
2.0 Background: The St. Paul ARS Cereal Disease Research Unit conducts research on fungal rust and Fusarium diseases of small grains to identify plant resistance genes, study pathogen populations, pathogen functional genomics, and study plant-pathogen interaction. An imaging system is needed to replace an imaging system that is no longer functioning and cannot be repaired. The imaging system is used to analyze DNA, RNA, and proteins on gels and membrane bolts to identify differences between samples and achieve experimental goals. The system also is used to examine fluorescently tagged proteins in fungi in plates/ dishes and in plants.
CONTRACTOR REQUIREMENTS
3.0 Technical Requirements/Tasks: Required specifications for an imaging system for chemiluminescent, RGB fluorescent, colorimetric detection of membrane blots, gels, plates, samples.
• Camera: 9.1 MP 16-bit, 65,536, Grayscale CCD, cooled to -50 ℃ relative, -20 ℃ absolute.
o Fixed motorized lens.
o Spatial resolution down to 40 µm (microns) and 60 µm (microns) o Ability to select no binning option (1x1) for high spatial resolution.
o Auto-focus o Camera distance to sample 24 cm (bottom tray)/ 15 cm (top tray)
• Imaging field of view 20.5 x 16.5 cm.
• Dual wavelength UV trans-illuminator, 302 nm and 365 nm.
• LEDs for gel and blot applications (blue 472 +/- 15 nm; green 524 +/- 12 nm;
red 628 +/- 16 nm).
• Multiplex two to three channel fluorescent detection (visible RGB).
• Chemiluminescent detection.
• Bioluminescence detection.
• True color marker imaging.
• True color imaging.
• Blue light fluorescent imaging.
• UV 302/365 imaging.
• Sample types to include: gels, membranes, plates, plants/ plant parts.

• Upgradable to EPI Laser modules at 685 nm and 784 nm for NIR applications.
• Light-tight cabinet for darkroom imaging.
• Pull-out, dual-wavelength UV trans-illuminator with 302 and 365 nm.
• Safety door sensor to prevent accidental exposure to UV and other excitation sources, which automatically deactivates light sources when the door is open unless safety override is activated.
• Safety tool for safety override.
• Safety override enables band cutting on the UV trans-illuminator, with automatic turn off time after 5 minutes for increased security (optional accessories for gel cutting are recommended for the best performance and ease of use).
• Two positions for imaging chemiluminescent and fluorescent samples:
bottom shelf for largest field of view and top shelf for highest resolution.
• Automatic detection of sample blot shelf position for automatic focus adjustment.
• Ability to configure to laptop computer.
• Ability to upgrade to analysis software.
• Ability to upgrade to trans-white table.

• Software to include:
o Automatic or user-specified manual imaging. Customizable protocols allowing users to specify and save imaging settings for repeated use.
o Gallery mode for file opening, editing, exporting, and saving in multiple formats; view and export images in multi-channel or single-channel visualization; image crop, flip, and rotation; image saving for publication at any DPI; image annotation tools; addition of notes to images.
o Preset automatic application modes (chemiluminescent blots, fluorescent labels, nucleic acids with dyes, protein gels with dyes, true color imaging) for fast optimized workflow; choice of autoexposure (adjustable RapidCapture, Wide Dynamic Range, Overexposure, or Extended Dynamic Range) and manual exposure for performance and flexibility.
o Fluorescent labels mode for up to 3 fluorescent channel imaging in a single acquisition.
o Chemiluminescent blot mode for detection of chemiluminescence with true color molecular weight marker workflow. Single, cumulative, and multiple exposure workflows.
o Nucleic acid mode enabling trans UV 302/365 and EPI Blue excitation and Visible Emission Detection; UV Override for band excision.
