Specifications Project: UPLC Analysis System Purchase for Vegetable Crops Research Unit
GENERAL INFORMATION
1.0 Scope of Work: The USDA-ARS is requesting quotes for purchase of a new Ultra
Performance Liquid Chromatography (UPLC) Analysis System and detectors (Diode array, refractive index, and mass spectrometer). Specifications for the purchase are itemized below.
2.0 Background: The USDA-ARS will use this system to collect data on composition of the crops investigated in the Vegetable Crops Research Unit. These crops include hemp, potato, cranberry, carrot, cucumber, and others. Compounds investigated will include cannabinoids, carbohydrates, pigments, and flavor compounds. Analyses will include both identification and quantification.
CONTRACTOR REQUIREMENTS
3.0 Technical Requirements/Tasks:
Flow path must be able to tolerate solvent pH from 2-12. The system will support leak sensing. System cycle time between injections shall be 30 seconds or less.
The pump must be a quaternary pump designed for Ultra Performance Liquid Chromatography, designed to blend up to four solvents simultaneously in any proportion or concentration. The pump shall have an integrated degasser, which operates up to the maximum flow rate, with an independent channel for each solvent. The degasser shall support an additional channel for the sample manager. The pump solvent composition must be settable from 0.0 - 100.0 % in 0.1 % increments within a single method. The maximum operating pressure of the pump must be 15,000 psi or 1,034 bar with user selectable upper and lower pressure limits within the settable range of the flow rate. The pump shall support automated priming of all four (4) solvent lines, from either a wet or dry state.
The sample manager must be designed for Ultra Performance Liquid Chromatography -able to operate up to 15,000 psi (1,034 bar) pressure. The sample manager contribution to dwell volume shall typically be < 20 μL default configuration (no loop installed). The sample manager must be a programmable, automatic liquid sampler for use in Ultra Performance Liquid Chromatography, Ultra High Performance Liquid Chromatography, or High Performance Liquid Chromatography. The sample manager must include sample preparation functions and auto dilution. The sample manager must be able to accommodate at least one hundred (100) of the standard 2-mL vials or >2 microplates (92 or other wellplates). The sample manager must have a settable temperature range from 4.0°C to 40.0°C in 0.1°C increments to provide an environment for temperature sensitive samples.
The sample manager must be able to perform replicate injections from 1 to 99 and shall have random access to any vial for multi-method operation.
The column heater compartment must support one column, from 1.0mm to 4.6mm ID to up to 150mm in length, with a pre-filter or guard column. The column heather compartment must have a temperature accuracy of less than ±0.5°C without the need to calibrate.
The refractive index detector must have the ability to set the sampling/data rate and filter time constants independently to maximize the resolution and sensitivity. The refractive index detector must be able to acquire data at up to eighty (80) points per second. The refractive index detector must have a linear dynamic range with less than 5% deviation over ± 5 x 10-4 RIU. The refractive index detector must have a flow cell with an internal volume of 1.3μL or less. The refractive index detector must have an internal volume of less than 15μL (measured at the detector inlet to the outlet of the reference cell and includes all internal fluidics to the detector).
The photodiode array detector must have standard analytical flow cell with a 10mm path length and a volume of not more than 500nL for optimal peak dispersion of UPLC separations. The photodiode array detector must have optional flow cells of both longer (25mm) and shorter (3mm) path length for optimizing detection of a wide range of applications. The UPLC system software must allow users to retrieve UV/Vis spectra from a defined peak and save it in a created spectral library for identification of compounds by matching with those of standards. The photodiode array detector must have a means to optimize lamp performance automatically and without user intervention. This compensates for lamp degradation over time without the need to frequently replace the lamp. The photodiode array detector must be able to acquire data at up to eighty (80) points per second. The photodiode array detector must have a noise specification of less than 6 μAU The photodiode array detector drift specification must be less than or equal to 1x10-3 AU/hour/°C
The mass spectrometer instrument will be a high-performance quadrupole mass analyzer. It shall be equipped with an XS Collision Cell where the collision energy is under programmable software control. The collision energy and collision gas pressure used during data acquisition shall be automatically recorded and appended to the relevant data file. The instrument shall incorporate an oaToF mass analyzer with a mass range up to m/z 100,000 and a resolving power of 40,000 FWHM.
