1. Must be able compatible with VRC’s established sequencing protocols, such as DNA/RNA library prep and data analysis, which have taken a substantial amount of time and resources to validate.

2. Must have the highest sequencing quality data: >85% Q30 and >90% Q30 quality scores for 2x50 bp and 2x150bp, respectively.

3. Must have eight (8) flow cell options, ranging from 100M to 1.2B reads and 30 to 360 Gb.

4. Must incorporate Dry Instrument Design: all reagent fluidics are contained in the disposable reagent cartridge, resulting in zero run-to-run contamination, less maintenance and no washes.

5. Must have an On-board DRAGEN Server, or equivalent: on-board BCL to FASTQ conversion, lossless data compression, and secondary analysis pipelines for RNA-Seq, differential expression, enrichment, scRNA-Seq and others.

6. Must have a small footprint: 60 x 60 x 65 cm (HxWxD) or 23.6” x 23.6” x 25.6”

4. Demonstrate that the nature of the product/service and the proposed contractor requires use of the authority cited.

a. Exclusive and Required Nature of the Product(s) and/or Service(s)

This instrument must have the following characteristics and must be compatible with our current instruments and sequencers. Also, the kits and reagents must be specific to the other machines that we have in our lab. We have used compatible sequencers and reagents in the past and changing variables in the middle of experiments would compromise the integrity of our results. The instrument must utilize innovative technologies and revolutionary assays to the analysis of genetic variation and function. By having these characteristics, the VRC will reduce time delays due to out-sourcing of sequencing and overall reduce cost to the government by providing faster results that will help aid in the design and generation of effective vaccines.

Also, these are characteristics that must be met in order to fulfill our need here at the VRC:

There is also an immediate need for this instrument. We are currently working around the clock on SARS CoV-2 and now Monkeypox. Given the nature of both of these viruses, any delay in acquisition of this instrument will negatively impact our critical research efforts.
