Statement of Work (SOW)

Project: Dissociation of tissue to obtain single-cell suspensions for genome editing

GENERAL INFORMATION

1.0 Scope of Work: The overall goal is to phenotype genome edited cells to identify and promote improved traits in pigs. Genome editing to produce changes in DNA that are thought to enhance production agriculture traits will be analyzed by culturing edited cells in vitro to determine if there is a positive effect on cell phenotype. The objective of this work is to be able to obtain single-cell suspensions from tissue that can be used to modify DNA from a single cell suspension.

2.0 Background: Scientists will conduct research to develop pigs with improved traits of interest for the swine industry by using genome editing techniques. The scientist will use genomic information to predict DNA sequence changes that will cause beneficial changes in pig production (increased muscle growth, cell proliferation, improved digestibility, disease resistance, etc.) and to test those changes, in vitro phenotyping of cells and organoids will be completed. To obtain cells with DNA edits, single cells must be able to be dissociated from tissue and used for genome editing. After genome modifications are created in the cells the subsequent change in phenotype will be validated in vitro before moving forward to produce a genome edited pig. The project requirement is for a DNA and protein gel imaging system that will allow us to analyze and validate whether the gene or protein is modified the way we expect and to measure the effects of genome editing on downstream molecular pathways and off target effects.

o Must be able to dissociate tissue into single cell suspensions o Must allow homogenization of tissues o Up to 8 samples should be able to be completed at a time
