DESCRIPTION: The National Institutes of Health- NIAAA intends to procure the services of a contractor to provide a GeoMx Digital Spatial Profiler. This instrument will be used in multiple NIAAA labs that are interested in spatial transcriptomics approach. The nanostring GeoMx Digital Spatial Profiler is a novel digital technology (proprietary to NanoString) that is based on multiplexed measurement of protein and nucleic acids and offers unparalleled levels of precision coupled with the ability to quantify up to 96 proteins and over 18,000 RNA targets on a formalin-fixed, paraffin-embedded (FFPE) or a fresh frozen (FF) tissue section on a microscope slide. This technology will allow the Lab to expand their technical capabilities to conduct spatial biology, which will uncover critical pathologic processes, eventually may help to identify therapeutic targets in multiple disorders. Using this technology will allow the to analyze detailed pathologic pathways analysis in as shorter time, with minimum sample requirement and cost-effective manner. Currently the Lab has already a Nanostring nCounter Max analyzer, which is the digital anaylzer of the GeoMx Digital Spatial profiler. Therefore, the new aquation will also work in synchrony with the existing system. Besides, combined of these system could detect fragmented nucleic acid samples from formalin fixed parafilm embedded histological tissue samples. This could give flexibility to use with human archive samples to make clinical relevance of the preclinical findings This system will help to minimize overall consumable cost for individual gene expression analysis. Accordingly, it will be time and cost effective. Furthermore, it will be used by multi-lab users and by multiple lab members including LPS and SFD in multiple projects. The system will allow them to perform detailed analysis in a shorter time and cost effective manner compared to the traditional approaches such as quantitative PCR methodology.

Based on NanoString’s proprietary digital barcoding technology, the GeoMx DSP platform measures local protein levels, and can be combined with RNA expression, within heterogeneous tissue samples. Combining both multiplexed nucleic acid and protein on the same platform gives researchers the ability to spatially resolve RNA when suitable antibodies do not exist. The GeoMx DSP assay is performed on the GeoMx DSP platform which includes imaging and fluidic components to capture spatial context, and current nCounter® or next-generation sequencing (NGS) instruments provide the quantification. Protein detection is enabled via primary antibodies which are covalently attached via a UV photocleavable linker to DNA indexing oligos. Following antigen retrieval, FFPE tissue samples are stained with a multiplexed cocktail of labeled antibodies, and DNA oligos are subsequently released by UV light exposure across regions of interest. The liberated DNA oligos are then hybridized to optical barcodes for quantitation on an nCounter or NGS instrument. This technique enables quantitative, multiplexed protein detection up to 5.5 logs of dynamic range.

The nCounter GeoMx Digital Spatial Profiler and associated reagents required to operate the system are only available from NanoString Technologies, Inc. Therefore, NanoString Technologies, Inc is the sole source for the requested nCounter GeoMx DSP system.

It is the intent of the National Institutes of Health (NIH)- NIAAA under the authority of FAR 6.302-1 to procure this equipment on a sole source to be able to continue and compare data from previous research using the very same equipment so that they can understand and treat alcohol disorders while developing appropriate models of addiction.

While NanoString’ system is not a unique “analyzer”, the program requesting this unit already has components and/or software compatible with the current equipment in their different Labs and would like to keep building on similar compatible instruments to save on maintenance.

The nCounter GeoMx Digital Spatial Profiler and associated reagents required to operate the system are only available from NanoString Technologies, Inc.

This is a notice of intent to award a sole-source, fixed price purchase order to NanoString Technologies Inc., located in Seattle, WA 98109 for commercial items prepared in accordance with FAR Parts 12 and 13.

It is the intent of the National Institutes of Health (NIH)- NIAAA under the authority of FAR 6.302-1 to procure this equipment on a sole source to be able to continue and compare data from previous research using the very same equipment so that they can understand and treat alcohol disorders while developing appropriate models of addiction.

In order to comply with the requirements in this Special Notice # NICHD-22-0103, the responses should be based on our interpretation of the general requirements provided above.

The proposed contract action is for supplies for which the Government intends to solicit and negotiate with only one source under the authority of FAR 6.302. However, interested vendors and 3rd party distributors may identify their interest and capability to respond to the requirement or submit quotations. This notice is not a request for competitive quotes but all quotes received within the time and date of publication of this " Notice” will be considered by the Government.

A determination by the Government not to compete with this proposed contract based upon responses to this notice is solely within the discretion of the Government. Information received will normally be considered solely for the purpose of determining whether to conduct a future competitive procurement.

HOW TO RESPOND: In order to compete for this project, interested parties must be registered in the Central Contracting Registration/SAM and submit a price quotation by 9:00 am, ET on May 30, 2022. Offerors should submit one copy of the quote to MOE FILALI via email.

Electronic transmissions to Moe.Filali@nih.gov are preferable but must be timely. It is the responsibility of the Offeror to ensure the proposal is received by the date and time shown above.

This is a “special Notice”/solicitation for commercial items prepared in accordance with the format in FAR 12.6 as supplemented with additional information included in this notice. This announcement constitutes the only solicitation and a separate written solicitation will not be issued. This solicitation number is NICHD-22-0103 issued as a Request for Quotation (RFQ).

The solicitation/contract will include all applicable provisions and clauses in effect through Federal Acquisition Circular 2019-05.
