Statement of Work Project: Real-Time PCR Replacement

GENERAL INFORMATION

1.0 Scope of Work: Purchase a Real-Time FAST capable PCR thermocycler to replace one of the machines currently in operation.

2.0 Background: The current ABI 7500 FAST Real-time PCR thermocyclers in the

Virus & Prion Disease Research Unit’s possession have been workhorses, producing massive amounts of data from animal samples for the past 15+years.

These machines are consistent and reliable. It is imperative to our scientific mission that we maintain continuity with our data in the future. A replacement machine must perform as similarly as possible and must have software compatible with 7500 v2.3 or higher as we have past, current, and future data that will be processed together. In addition, we have fully stocked with the consumable supplies needed for use with our current machine.

CONTRACTOR REQUIREMENTS

3.0 Technical Requirements/Tasks:

o Thermocycler shall have Peltier-based thermal cycling system.

o Thermocycler shall use 96-well plate format, with the ability to run 8-well strips.

o Thermocycler shall use Reaction Volume of 10-30μL and shall be compatible for use with 0.1mL reaction plates.

o Thermocycler shall be calibrated for dyes TAMARA, Cy3, FAM, SYBR Green, Texas Red, JOE, ROX, Cy5, NED, VIC.

o Thermocycler shall have 9 logs of linear dynamic range.

o Thermocycler shall have the ability to distinguish between 5,000–10,000 genome equivalents (two-fold copy number difference) with 99.7% confidence.

o Thermocycler shall perform with a sample ramp rate of ± 5.5°C⁄sec with a temperature range of 4.0-99.9°C.

o Thermocycler shall have compatibility with Fast Cycling chemistries &

TaqMan Gene Expression Assay o Dimension of unit shall not exceed 13”wide x 17”deep x 19”tall. This must be a table-top unit.

o Thermocycler shall include stand-alone computer interface with applicable software for data analysis (compatible with 7500 v2.3 or higher) o Onsite installation by OEM representative shall be included with quote.

o Qty: 1

*Trade-in credit required. We will be trading the following unit currently in use:

Applied Biosystems 7500FAST Real Time PCR machine (SN 275012920), >10yrs old, good condition. Any credit for this trade-in shall be included with quote.

4.0 Government Furnished: Installation representative will require access to USDA-

NADC, Building 20, Laboratory 2728.

5.0 Deliverables/Schedule: Unit shall be delivered onsite within 30 days of date of award, with installation completed within 45 days of date of award.

6.0 Evaluation Factors: Lowest Price technically acceptable.

7.0 Technical Point of Contact: Should there be technical questions, please contact

Sarah Anderson at 515-337-7036, sarah.j.anderson@USDA.gov.

8.0 Place of Delivery: USDA, 1920 Dayton Avenue, Bldg 24, Ames, IA 50010
