The Trustees of the University of Pennsylvania, doing business as Clinical Practices of the University of Pennsylvania, was awarded a $495,024 federal project grant from the National Institute on Drug Abuse (CFDA 93.279 - Drug Use and Addiction Research Programs) to study the effects of psilocybin on brain mechanisms, cognitive flexibility, and motivated behaviors in individuals with opioid use disorder.

The 2-year grant project will prospectively assess 72 participants, with 24 in the initial R61 phase and 48 in the subsequent R33 phase. The research aims to examine psilocybin's impact on motivational circuitry, inhibitory control, and neurocognitive flexibility in this population. Exploratory measures will also evaluate psilocybin's effects on craving, withdrawal, depression, and prior adversity. Findings from brain-behavioral probes showing large effect sizes in the R61 phase will then be further examined through network-level brain changes in the R33 expansion.

This study is intended to provide critical mechanistic insights to support the rational advancement of psilocybin as a potential treatment for opioid use disorder and other disorders with unmet clinical needs.
