The National Institute on Aging (NIA), under the Aging Research Federal Grant Program (CFDA 93.866), awarded a $254,250 Project Grant (R21AG087904) to New York University School of Medicine to develop a multi-scale model of brain aging and Alzheimer's disease (AD) using advanced diffusion MRI (dMRI) techniques. The project aims to create a brain microstructure age framework by generating brain charts of quantitative dMRI and mesoscopic MRI (meso-MRI) markers across the lifespan. It will evaluate the effects of race/ethnicity, sex, and lifestyle factors on brain aging, as well as the sensitivity of brain microstructure to increased aging, neuroinflammation, and neurodegeneration along the AD/dementia continuum. The research will be validated using postmortem brain tissue analysis and proteomics. The goal is to achieve a clinically feasible, highly sensitive brain age framework that provides insights into the microstructural processes underlying brain aging and AD/dementia, enabling quantitative assessment of treatment responses.
