This federal Project Grant award of $525,436 from the National Institute of Neurological Disorders and Stroke (NINDS), under the Extramural Research Programs in the Neurosciences and Neurological Disorders (CFDA 93.853) program, will support research to elucidate the molecular mechanisms underlying the loss of potassium selectivity and constitutive activation observed in Keppen-Lubinsky Syndrome (KLS) disease mutations in the GIRK2 potassium channel. The research team at the Icahn School of Medicine at Mount Sinai will use a multi-disciplinary approach combining structural biology, computational modeling, and human neuron cell models to identify and characterize novel selective inhibitors of the KLS GIRK2 channels. This work aims to develop a foundation for a potential new treatment for the severe neurological symptoms associated with KLS, a rare genetic disorder characterized by developmental delays, intellectual disability, and neuromuscular abnormalities.
