The recipient will determine the influence of changing glutathione ratios (GSH:GSSG) on protein glutathionylation driving MERTK signaling in efferocytosis, the process by which macrophages clear apoptotic cells and resolve inflammation. Research activities include in vitro studies using human cells and macrophage cultures isolated from glutathione synthesis mutant mice to characterize how glutathionylation disrupts efferocytosis, and in vivo studies in murine models with mutations in the GSH synthesis pathway on high-fat diet to drive atherosclerosis. The work will test the hypothesis that decreased GSH:GSSG ratios impair efferocytosis and promote plaque instability, and will explore therapeutic approaches to restore defective efferocytosis by stimulating a more reductive cellular environment. Work is performed in Shreveport, Louisiana. The period of performance runs from July 15, 2026, through April 30, 2031. The assistance type is a Project Grant.
