The research expands preliminary investigations into cerebellar Purkinje cell activity and rapid-eye-movement (REM) sleep regulation. The project will test whether cerebellar activity is sufficient to modulate REM sleep, anatomically and electrophysiologically characterize the Purkinje cell to brainstem pathway enabling cerebellar modulation of REM, and examine two mouse models—tottering mice (episodic ataxia type 2) and Purkinje-cell specific TSC1 knockout mice (autism)—to determine whether cerebellar dysfunction leads to sleep dysfunction. The work will employ an innovative longitudinal monitoring system to continuously track sleep state and actigraphy across 24/7 observation periods of up to lifelong duration in these models. The research aims to mechanistically link cerebellar dysfunction to REM sleep disturbance and explain comorbidity patterns observed in cerebellar disorders including spinocerebellar ataxias, Friedreich's ataxia, and ataxia telangiectasia.
