The National Heart, Lung, and Blood Institute (NHLBI) awarded a $678,803 Project Grant (CFDA 93.837 Cardiovascular Diseases Research) to The Ohio State University to conduct research on the role of the mitochondrial deubiquitinase USP30 in regulating endothelial cell function and miRNA biogenesis, and its potential as a therapeutic target for acute inflammatory diseases like acute respiratory distress syndrome (ARDS) and sepsis.
The research aims to: 1) determine if inhibiting endothelial USP30 can reduce inflammation and preserve endothelial barrier function in experimental models of lung injury; 2) elucidate the molecular mechanisms by which USP30 inhibition modulates MAT2A stability, S-adenosylmethionine production, and miR-30a-5p expression; and 3) assess whether the protective effects of USP30 inhibition are mediated through modulation of miR-30a-5p. The project will run from February 1, 2025 through November 30, 2028.
