The University of California, San Diego (UCSD) was awarded a $639,898 Project Grant (R01HG014094) from the National Human Genome Research Institute (CFDA 93.172 - Human Genome Research) to develop a multimodal MPRA platform for studying the mechanisms and grammar of regulatory sequence function.

The overarching goal is to create new technology based on massively parallel reporter barcoding strategies that can simultaneously measure the effects of sequence perturbations on chromatin state, DNA-protein interactions, 3D chromatin interactions, and transcription regulation at promoters. This will enable the researchers to test the hypothesis that individual enhancers may exert their function through modulation of different transcriptional mechanisms, such as initiation versus pause release, during cellular differentiation or acute stimulus responses. The new methodology will be validated against genome editing of endogenous loci. The project period runs from January 1, 2025 to December 31, 2028.
