The research addresses glomerulosclerosis and tubulo-interstitial fibrosis in chronic lupus nephritis, pathogenic aspects insufficiently addressed by current therapies and strongly associated with progression to end-stage renal disease. The project tests the hypothesis that syndecan-1, induced by inflammation, modulates PTPRS activity on plasmacytoid dendritic cells, thereby promoting inflammation and fibrosis. Three aims assess the regulation of PTPRS by syndecan-1 in lupus nephritis and plasmacytoid dendritic cells; identify PTPRS substrates and molecular mechanisms in these cells; and evaluate PTPRS's role in inflammation-fibrosis cross-talk and disease chronicity. The period of performance runs from September 1, 2026, through May 31, 2030, with work performed in West Hollywood, California.
