The National Cancer Institute (NCI) awarded a $680,334 Project Grant (CFDA 93.396 - Cancer Biology Research) to the Icahn School of Medicine at Mount Sinai (ISMMS) to conduct research on the cellular and molecular mechanisms driving age-associated pathogenic myelopoiesis and its contribution to chronic inflammation and tumor progression. The 5-year project aims to: 1) identify the cellular players involved in the IL-1 signaling response and their role in age-associated inflammation and cancer; 2) define the central role of DNA methyltransferase 3A in age-dependent changes to the myeloid cell compartment; and 3) characterize how age-associated reactivation of transposable elements in myeloid cells contributes to age-related inflammation. This research seeks to elucidate the specific molecular mechanisms and cellular pathways driving the link between immune aging, myeloid dysregulation, and increased cancer risk in older populations. The results are expected to enable the development of new therapeutic approaches targeting pathogenic myelopoiesis and associated inflammation in aged patients.
