This Project Grant award from the National Cancer Institute (NCI), under the Cancer Treatment Research program (CFDA 93.395), will support research at Cold Spring Harbor Laboratory (CSHL) to optimize anti-NMDAR antibodies for the treatment of triple-negative breast cancer (TNBC). The key products and services to be delivered include: Establishing an orthotopic mouse model of NMDAR-positive TNBC to enable the isolation and characterization of NMDAR-reactive antibodies through single-cell RNA sequencing of intratumoral B cells (Aim 1). The project will then use cryo-EM to screen these antibodies for structural changes that may be associated with neurotoxicity (Aim 2). Finally, candidate antibodies predicted to have low pathogenicity will be evaluated for therapeutic efficacy and toxicity in the NMDAR-positive TNBC mouse model (Aim 3). The total award amount is $781,436, spanning the project period from April 14, 2025 to March 31, 2030. This research aims to establish a safe and effective anti-NMDAR antibody therapy for the approximately 40,000 TNBC patients diagnosed annually in the United States, addressing a critical unmet need.
