The National Cancer Institute (NCI) has awarded a 5-year, $523,292 Project Grant (CFDA 93.393 - Cancer Cause and Prevention Research) to the Dana-Farber Cancer Institute, Inc. (DFCI) to support research aimed at exploiting a novel DNA repair defect in Ewing sarcoma, an aggressive form of bone and soft tissue cancer. The primary goals of the project are to: 1) investigate the molecular mechanisms by which the microhomology-mediated end-joining (MMEJ) DNA repair pathway is suppressed in Ewing sarcoma; 2) assess the function of the MMEJ pathway in Ewing sarcoma primary tumor tissue from patients; and 3) explore therapeutic approaches to exploit the MMEJ defect for the benefit of Ewing sarcoma patients. The research conducted under this grant will advance the understanding of DNA repair abnormalities in Ewing sarcoma and provide insights into potentially novel treatment strategies targeting this unique cancer-specific DNA repair defect.
