The research builds upon a completed Phase I clinical trial of an adenoviral vaccine (NOUS-209) containing 209 shared mutated neoantigens derived from mismatch repair-deficient tumors. The project will conduct immunological studies examining T cell receptor (TCR) clonotypes against shared mutated neoantigens in colorectal mucosa, testing the central hypothesis that specific memory T cells can be expanded through neoantigen vaccination to elicit adaptive anti-tumor immunity in at-risk Lynch Syndrome carriers.
