The award funds a study at the University of South Alabama examining how the carbonic anhydrase IX isoform and cytotoxic amyloid proteins impact pulmonary endothelial cell function during Pseudomonas aeruginosa infection. Specifically, the research will test the hypotheses that CA IX is critical to acid regulation and barrier integrity in pulmonary microvascular endothelial cells, and that P. aeruginosa elicits amyloid production causing CA IX shedding and increased lung injury. In vitro experiments using genetic approaches and cell assays will evaluate acidosis effects and specific CA IX domain roles under physiologic and infectious conditions. In vivo and ex vivo studies using mouse models of acidosis, pneumonia and isolated lung perfusion will translate findings. Successful completion could provide new insights into acidosis mechanisms in pneumonia to identify CA IX and amyloids as biomarkers and therapeutic targets, advancing the goals of the Cardiovascular Diseases Research program (CFDA 93.837) supporting heart and vascular research and training.
