UNC-CH's Axtman Lab will design and synthesize proteolysis targeting chimeras (PROTACs) for TDP-43, exploring the location of linker attachment and the nature of the linker. They will then evaluate the PROTACs to determine if they can induce degradation of TDP-43 in immortalized cells and iPSC-derived neurons. This work will help inform the iterative development of new PROTAC molecules and assess the biological consequences of TDP-43 degradation, with the goal of advancing a novel therapeutic approach for ALS.
