The research team will generate in vitro and in a reconstituted synthetic biology system a "carbohydrate chaperone"—a protein complex that stimulates crystallization of branched glucan polymers. The proposed glucan chaperone consists of ISA1 and ISA2 proteins in a heterotetrameric complex with an elongated structure containing eight glucan binding sites organized to bring linear chains into proximity and stimulate formation of double helices. The non-mutant complex also catalyzes branch linkage hydrolysis. Secondary school students will perform structural design of proteins for the experimental system. Discoveries from the experimental system could impact biotechnological applications of polymer science. Performance occurs in Ames, Iowa from September 1, 2026 through August 31, 2029.
