The National Science Foundation Division of Chemistry awarded Clemson University a $492,000 Project Grant under the Mathematical and Physical Sciences federal grant program (CFDA 47.049) to study mechanisms of antifungal resistance. Professors Julia Brumaghim and Lukasz Kozubowski at Clemson University will investigate how azole antifungal compounds interact with metals like copper and iron to generate reactive oxygen species that damage fungal DNA and lead to chromosomal instability and drug resistance. The research aims to test the hypothesis that metal binding and reactive oxygen species generation by azoles underlie the development of resistance. Key activities will include quantifying how diverse azoles bind metals and promote oxidative DNA damage in vitro, and determining their effects on reactive oxygen species, DNA integrity and resistance development in the model fungi Saccharomyces cerevisiae and Cryptococcus neoformans under normal and elevated metal conditions. Results of this work have the potential to advance understanding of antifungal resistance mechanisms and guide future antifungal development.
