The National Cancer Institute (NCI) has awarded a $1,390,720 Cooperative Agreement under the Cancer Treatment Research program (CFDA 93.395) to The Leland Stanford Junior University (Stanford University) to develop an enhanced chimeric antigen receptor (CAR) T-cell therapy targeting the GPC2 antigen for treating medulloblastoma, a common and deadly pediatric brain cancer.

The project aims to optimize the GPC2-CAR T-cell therapy by overexpressing the c-Jun transcription factor, which has been shown in preclinical studies to improve the potency and persistence of the CAR T-cells against medulloblastoma and neuroblastoma tumors. The third aim will then apply this process to produce clinical-grade GPC2.c-Jun-CAR T-cells for evaluation in a Phase 1 trial assessing the feasibility, safety, and efficacy of this potency-enhanced CAR T-cell therapy against medulloblastoma. This project leverages Stanford's deep expertise in CAR T-cell engineering and will provide valuable data on GPC2 as a target for medulloblastoma treatment.
