INTRODUCTION PURSUANT TO FAR Subpart 5.2-Synopses of Proposed Contract Actions, THIS IS A PRE-SOLICITATION NOTICE OF A PROPOSED CONTRACT TO ACTION. THIS IS A PRE-SOLICITATION NON-COMPETITIVE NOTICE OF INTENT TO AWARD A CONTRACT OR PURCHASE ORDER WITHOUT PROVIDING FOR FULL OR OPEN COMPETITION (INCLUDING BRAND-NAME). The Office of Acquisition (OA) at the National Institute on Drug Abuse (NIDA) at the National Institutes of Health (NIH) under the Department of Health & Human Services (DHHS), on behalf of the National Center for the Advancement of Transalational Sciences (NCATS) intends to negotiate and award a contract to obtain contractor services for proteomic analysis of minibrains and cortical glutamatergic neurons by Poochon Scientific LLC located at 8435 Progress Drive, Frederick, MD 21701. The determination by the Government to award a contract without providing for full and open competition is based upon consistency with prior data results and continuity of science and on market research conducted. Specifically, the sources sought type of notice under this solicitation, released on FedBizOpps from January 30, 2019 tough February 11, 2019, indicates only one source of supply or service is capable of achieving this Government requirement. NORTH AMERICAN INDUSTRY CLASSIFICATION SYSTEM (NAICS) CODE The intended procurement is classified under NAICS code 541380 Testing Laboratories with associated small business size standard of 15,000,000 USD. REGULATORY AUTHORITY The resultant contract will include all applicable provisions and clauses of the Federal acquisition Requlation (FAR) in effect tough the Federal Acquisition Circular (FAC) 2019-01 / 01-22-2019. STATUTORY AUTHORITY This acquisition is conducted under the authority of the Federal Acquisition Regulation (FAR) Part 13-Simplified Acquisition Procedures, Subpart 13.106-1 (b) (1), Soliciting from a single source. DESCRIPTION OF REQUIREMENT Project Description Statement of Work: The Contractor shall perform the following statement of work: Contractor shall receive frozen brain tissue samples or frozen cortical glutamatergic neurons from NCATS. Contractor will provide the following services: protein lysate preparation, TMT-labeling, UHPLC fractionation, digestion, LC-MS/MS analysis, TMT quantitation, pathway analysis and report of results. Contractor shall use PROMIC and PROMICPATH for the required services. Independently and not as an agent of the Government, the Contractor shall furnish all the necessary services, qualified personnel, material, equipment, and facilities, not otherwise provided by the Government as needed to perform the following tasks. Specifically: 1. Sample preparation including: a. Protein lysate preparation from 45 samples. b. TMT-labeling. c. UHPLC fractionation and digestion 2. LC-MS/MS analysis: a. TMT10plex-Labeling Quantitative Proteome Profiling b. Quantitatively comparison of the proteomes (>6000 proteins) of 7-10 samples in one set of analysis using Thermo TMT10plexTM Isobaric Label Reagent. c. Nanospray LC/MS/MS analysis and database search 3. Data analysis and report: a. TMT quantitation analysis b. Pathway analysis c. Result report Period of Performance Delivery of results will be proived in eight (8) weeks from contractor receipt of material. GOVERNMENT RESPONSIBILITIES Government will have the responsibility to review and approve the final report. Government will have the responsibility to provide frozen samples in order for the contractor to accomplish the necessary work. The Government will provide the cell pellets from the patient-derived induced pluripotent stem cells. The Government is responsible for sample/package preparation. DELIVERY OR DELIVERABLES Contractor will provide the data analysis and report for the contents of this statement of work. The data package will include a spreadsheet of raw data as well as a complete data work-up and full report. Offerors should indicate an estimated delivery time in full after receipt of an order and any other training or warranty considerations applicable. Inspection and acceptance will be performed at NCATS located at 9800 Medical Center Drive, Rockville, MD 20852. Purpose and Objectives: The purpose of this requirement is to obtain contractor results of proteomic analysis of minibrains and cortical glutamatergic neurons using TMT-multiplex labeling and LC/MS/MS. This order provides for testing results of an analysis to identify and compare proteomic changes in minibrains and cortical glutamatergic neurons. Proteomic analysis of minibrains and cortical glutamatergic cells using TMT-10plex labeling and LC/MS/MS is required to support work on N-glycanase deficiency (NGLY1), a rare genetic disease resulting from a deficiency in the N-glycanase-1 enzyme. The Proteomic analysis will be used to identify different protein expressions (proteomes) and compare minibrains and neurons with control cell lines, using TMT-10plex labeling and liquid comatography-tandem mass spectrometry (LC/MS/MS) based quantitative proteomics. Proteomic profiling will be performed for 24 brain tissue samples from 8 groups and 21 cortical glutamatergic neurons using the Thermo TMT100plex isobaric label reagent. The service includes protein lysate preparation, TMT-labeling, UHPLC fractionation, digestion, LC-MS/MS analysis, TMT quantification, pathway analysis and associated reports. CLOSING STATEMENT This synopsis is not a request for competitive proposals. However, interested parties may identify their interest and capability to respond to this notice. Responses to this solicitation must include clear and convincing evidence of the offeror's capability of fulfilling the requirement as it relates to the technical evaluation criteria. The price proposal must include the labor categories, an estimate of the number of hours required for each labor category, fully loaded fixed hourly rate or each labor category, breakdown and rationale for other direct costs or materials, and the total amount. In addition the Dun & Bradstreet Number (DUNS), the Taxpayer Identification Number (TIN), and the certification of business size must be included in the response. All offerors must have an active registration in the System for Award Management (SAM) at www.sam.gov. A determination by the Government not to compete this proposed contract based upon responses to this notice is solely within the discretion of the Government. The purpose of this potential requirement is to obtain testing results of an analysis to identify and compare proteomic changes in minibrains and cortical glutamatergic neurons. Project Requirements This Small Business Sources Sought notice seeks responses from qualified sources that are capable of providing the following salient service requirements of this potential government requirement deemed as the essential features needing to be achieved. Specifically, Offerors to this Small Business Sources Sought notice shall possess the capability to provide the following: Contractor will receive frozen brain tissue samples or frozen cortical glutamatergic neurons from NCATS. Contractor will provide the following services: protein lysate preparation, TMT-labeling, UHPLC fractionation, digestion, LC-MS/MS analysis, TMT quantitation, pathway analysis and report of results. Contractor shall use PROMIC and PROMICPATH for the required services. NCATS will ship the frozen samples to Contractor via FedEx overnight shipping on dry ice. The Government is responsible for sample/package preparation. The NCATS point of contact (POC) will inform Contractor when cells have been shipped out. Upon receiving samples, Contractor will inspect the vials and ensure samples have arrived in proper conditions and will take necessary steps for proper storage of samples until project begins. The data package will include a spreadsheet of raw data as well as a complete data work-up and full report. Reporting Requirements: 1. Within two (2) business days of completion of the LC-MS/MS analysis, Contractor shall inform the NCATS POC of the completion. 2. Contractor shall provide summaries and documentation to NCATS as necessary. Regular meetings between NCATS and Contractor will be scheduled either via teleconference or email. Contractor shall provide technical consultation and support at NCATS staff meetings as requested to facilitate ongoing communication. 3. Contractor will provide the data analysis and report for the contents of this statement of work. Contractor will provide monthly updates on the progress of the project, or more often as appropriate. CAPABILITY STATEMENTS/INFORMATION SOUGHT Interested parties are expected to review this notice to familiarize itself with the requirements of this project. Respondents must provide clear and convincing evidence of possessing the knowedge, skills, abilities and experience to successfully perform the requirements described in this announcement. Detailed prior experience in implementing similar requirements to the requirement described in this announcement must be clearly delineated in any response provided. Respondents must provide clear and convincing documentation of their capability in providing analytical quality control and any other requirements and services specified in this notice. Respondents must provide a general overview of the respondents' opinions about the difficulty and /or feasibility of the potential requirement, and any information regarding innovative ideas or concepts. The Respondent must also provide information in sufficient detail of the respondents' (a) staff expertise, including their availability, experience, and formal and other training; (b) current in-house capability and capacity to perform the work; (c) prior completed projects of similar nature; (d) corporate experience and management capability; and (e) examples of prior completed Government contracts, references, or other related information.
