The Department of Health and Human Services (DHHS), National Institutes of Health (NIH), National Cancer Institute (NCI), Division of Cancer Epidemiology and Genetics (DCEG) has a requirement for external scientific input regarding the conduct and analysis of the Sherlock lung project, particularly with regard to analytic approaches and the creation of novel methods, tools, and techniques for analyzing genomic data and studying clonal evolution and lung cancer heterogeneity. This is a combined synopsis/solicitation for commercial items, prepared in accordance with format in FAR 12.6 as supplemented with additional information included in this notice. This announcement constitutes the only solicitation and a separate written solicitation will not be issued. This solicitation, 75N91019Q00035, is issued as a request for quote (RFQ) and includes all applicable provisions and clauses in effect tough FAR Federal Acquisition Circular (FAC) 2019-01/ January 22, 2019, simplified acquisition procedures for commercial items. The North American Industry Classification System code is 541990 and the business size standard is $15M. Only one award will be made as a result of this solicitation. This will be awarded as a firm fixed price purchase order for severable services. BACKGROUND The Department of Health and Human Services (DHHS), National Institutes of Health (NIH), National Cancer Institute (NCI), Division of Cancer Epidemiology and Genetics (DCEG) is conducting the Sherlock lung project, a large study of lung cancer in never smokers. Lung cancer in never smokers includes approximately 10-25% of all lung cancers worldwide, ranks among the most common causes of cancer mortality, and has a distinct natural history, restricted histological subtype (almost exclusively adenocarcinoma), likely different profile of oncogenic mutations, and response to targeted therapy compared to lung cancer in smokers. Although a few risk factors are known to contribute to the etiology of lung cancer in never smokers, a large fraction of cancer cases cannot be explained by established environmental and genetic risk factors, highlighting the need for research in this area. One promising approach to identify the etiological factors involved in lung tumorigenesis in never smokers is based on the study of the "mutational signatures" that the exogeneous and endogenous processes leave on the tumor tissue and surrounding areas. This approach has identified over 50 different mutational signatures in other cancer types to date, corresponding to several oncogenic processes. The primary goals of the Sherlock lung study are: (1) to identify mutational signatures of the endogenous and exogenous processes involved in lung tumorigenesis and order them along the lung tumor evolutionary trajectory, and (2) to develop an integrated molecular, histological and radiological classification of lung cancer in never smokers. Investigators will conduct an integrative genomic analysis of mutational signatures in tumors and surrounding non-tumor ("normal") lung tissue from 2,000 never smokers. The cases will include a subset of "special exposure cases" exposed to high levels of known risk factors for lung cancer (n=~500) and cases from the "general population" without known risk factors (n=~1,500). Cases from many geographical areas and ethnic groups will be included to study the contribution of different germline and environmental factors. The tumor/normal genomics analysis will include whole genome sequencing, RNA sequencing, and genome-wide methylation arrays. The integrated molecular landscape will be ordered along the evolutionary trajectory of the tumors to infer the cascade of events leading to tumor formation and progression. Data on the molecular and evolutionary landscape will be combined with data from the histological examination of H & E slides from multiple tissue blocks per tumor and related to CT-scan imaging to provide a more refined classification of lung cancers among never smokers. Additional analyses for subsets of the cases will include liquid biopsy, tumor microenvironment, clonal evolution, and mining of electronic medical records and national databases to look for associations with other medical conditions and conic medications. This comprehensive research program will contribute to the understanding of the etiology and progression of lung cancer in never smokers and will provide clues into prognosis and treatment strategies. OBJECTIVE Sherlock lung investigators are seeking external scientific input regarding the conduct and analysis of the Sherlock lung project, particularly with regard to analytic approaches and the creation of novel methods, tools, and techniques for analyzing genomic data and studying clonal evolution and lung cancer heterogeneity. (i) Taxes are considered delinquent if both of the following criteria apply: (A) The tax liability is finally determined. The liability is finally determined if it has been assessed. A liability is not finally determined if there is a pending administrative or judicial challenge. In the case of a judicial challenge to the liability, the liability is not finally determined until all judicial appeal rights have been exhausted. (B) The taxpayer is delinquent in making payment. A taxpayer is delinquent if the taxpayer has failed to pay the tax liability when full payment was due and required. A taxpayer is not delinquent in cases where enforced collection action is precluded. (ii) Examples. (A) The taxpayer has received a statutory notice of deficiency, under I.R.C. 6212, which entitles the taxpayer to seek Tax Court review of a proposed tax deficiency. This is not a delinquent tax because it is not a final tax liability. Should the taxpayer seek Tax Court review, this will not be a final tax liability until the taxpayer has exercised all judicial appeal rights. (B) The IRS has filed a notice of Federal tax lien with respect to an assessed tax liability, and the taxpayer has been issued a notice under I.R.C. 6320 entitling the taxpayer to request a hearing with the IRS Office of Appeals contesting the lien filing, and to further appeal to the Tax Court if the IRS determines to sustain the lien filing. In the course of the hearing, the taxpayer is entitled to contest the underlying tax liability because the taxpayer has had no prior opportunity to contest the liability. This is not a delinquent tax because it is not a final tax liability. Should the taxpayer seek tax court review, this will not be a final tax liability until the taxpayer has exercised all judicial appeal rights. (C) The taxpayer has entered into an installment agreement pursuant to I.R.C. 6159. The taxpayer is making timely payments and is in full compliance with the agreement terms. The taxpayer is not delinquent because the taxpayer is not currently required to make full payment. (D) The taxpayer has filed for bankruptcy protection. The taxpayer is not delinquent because enforced collection action is stayed under 11 U.S.C. 362 (the Bankruptcy Code). The technical evaluation will be a determination based on information furnished by the Contractor. The Government is not responsible for locating or securing any information which is not identified in the quotation. The Government intends to evaluate quotations and make a single award without conducting "discussions" as defined in FAR part 15. Therefore, the Quoter's initial quote should contain the Quoter's best terms from a price and technical standpoint. However, the Government reserves the right to request quote revisions that seek additional price discounts, provide revisions to the technical quote, or clarify aspects of the quote. In the event quote revisions are requested, the Government will not necessarily be seeking revisions from all Quoters. The Government may reject any or all quotes if such action is in the public interest; accept other than the lowest quote; and waive informalities and minor irregularities in quotes received. The Government will issue an order resulting from this request for quotation to the responsible Quoter whose quotation results in the best value to the Government, considering both price and non-price factors. The following factors will be used to evaluate quotations: Personnel Qualifications: The experience, education, and qualifications of personnel proposed to work on the contract will be evaluated to determine their ability to perform their proposed duties. Specifically, the Contractor will be evaluated on the ability to: Demonstrate expertise in the areas of bioinformatics, computational biology, modelling, and statistical genetics. Demonstrate experience leading research on cancer evolution and heterogeneity, analyzing whole genome sequencing data, and applying novel techniques and sequencing technology to these analyses. Demonstrate the ability to apply methods or tools, and to modify existing methods or tools as required to identify clonal and subclonal copy number aberrations, single nucleotide variants, structural variants, and other genomic changes. Technical capabilities: The Government will evaluate the Quoter's technical capabilities based on demonstrated relevant examples of organizational experience and expertise. The Government will also look at feasibility of methodologies and services provided for successfully completing the tasks listed in the Statement of Work. Relevant is defined as work similar in complexity and magnitude of the work described in this SOW. Past performance: The Government will evaluate the Quoter's past performance based on information obtained from references provided by the Quoter, other recent and relevant past performance information obtained from other sources known to the Government, and any information supplied by the Quoter concerning problems encountered on the identified contracts and corrective action taken. Recent is defined as within the last tee years. Relevant is defined as work similar in complexity and magnitude of the work described in this SOW. Past performance on related contracts will be evaluated to determine, as appropriate, successful performance of contract requirements, quality and timeliness of delivery of goods and services, communications between contracting parties, proactive management and customer satisfaction, etc. The Government will access the relevant risks associated with each Quoter. Performance risks are those associated with a Quoter's likelihood of success in performing the acquisition requirements as indicated by that Quoter's record of past performance. Price: The Government will evaluate the Quoter's price for all items/services proposed. Price quotations will be evaluated for reasonableness. For a price to be reasonable, it must represent a price to the Government that a prudent person would pay when consideration is given to prices in the market. Normally, price reasonableness is established tough adequate price competition. EVALUATION OF OPTIONS It is anticipated that a purchase order awarded as a result of this solicitation will contain option provision(s) and period(s). The Government will evaluate quotations for award purposes by adding the total price for all options to the total price for the basic requirement. To account for the option period(s) possible under 52.217-8 (maximum of six months), Option to Extend Services, the Government will evaluate the option to extend services (FAR Clause 52.217-8) by adding 6 months of the Quoter's final option period price to the Quoter's total price. This amount will be the total evaluated price. . 75N91019Q00035 Department of Health and Human Services National Institutes of Health
