Only one award will be made because of this solicitation. This will be awarded as a firm fixed price type contract.
BACKGROUND
The U.S. Department of Health and Human Services, National Institutes of Health, National Cancer Institute (NCI), Center for Cancer Research (CCR). The intent of this purchase is to acquire a microscope, for the purpose of obtaining very high-resolution data with extremely high sensitivity for the analysis of submicron extracellular vesicles (EVs). The purpose of the instrument is to be able to perform studies that will identify and characterize single EVs and single fluorescent molecules across multiple lasers. This ultra-high level of sensitivity and precision is required for tests that will detect and enumerate EVs that have as few as a single molecule of interest. The purpose of this instrument is to provide single-molecule resolution for EV analyses as part of the EV analysis pipeline and the characterization of nanomaterials that is integral to the research that is being conducted in the Translational Nanobiology Section of the Laboratory of Pathology.
The salient characteristics that apply to this acquisition are as follows:
• Single nanoparticle (>20 nm) detection
• Single fluorescent molecule detection across multiple fluorophores
• Single particle number concentration enumeration
• Permanent alignment
• Ability to perform all operations on a standard desktop without the need for a vibration- isolating optical table
• Autofocus system
• Class 1 laser safe device
• Microscope unit and light engine with less than 465 cm2 and 810 cm2 respectively in footprint * Real-time calibrated power readings at the sample plane
• Optional integrated microfluidics upgrade including built in software for programmed delivery
• Two modes of operation to achieve super-resolution: single-molecule localisation microscopy (SMLM) and multifocal structured illumination microscopy (mSIM)
• Resolution by SMLM xy: 20 nm; z: 50 nm
• Three modes of illumination: widefield, TIRF and HILO
• Simultaneous dual emission channels
• 50×80 μm homogeneous field of view
• Homogeneous top-hat illumination
• Advanced piezo stage driven in X, Y and Z with 1 nm steps and 30 nm reproducibility
• Real-time (GPU) localization and quantitative analysis
• Whole-body temperature control at 37°C
• sCMOS Hamamatsu orca flash 4 V3, with high quantum efficiency 82% and high full frame acquisition rate 100fps
• Unlimited software licenses. This includes unlimited lifetime upgrades and free licenses for data analysis
• A minimum of 12-month manufacturer warranty. Online support for the first 12 months after commissioning, if needed.
• Must include training and installation
• Quote must include shipping, including (FBO)and installation cost. NCI will not be responsible for any cost not included.
Note: NCI plans to award a contract that has FOB origin delivery terms.
PAYMENT
Payment shall be made upon the delivery, installation, acceptance and completion of training.
DELIVERY / INSTALLATION
The Contractor shall deliver and install the required item(s) within 30 days of purchase order award to the following location:
10 Center Drive
10CRC
Bethesda, MD 20892
The Contractor that receives the award will be provided with the complete mailing address and point of contact. Upon award, contractor must notify the NCI Technical Point of Contact to schedule the delivery.
