About this file
This statement of work outlines requirements for single cell genomics reagent kits to support research at the USDA Agricultural Research Service Animal Parasitic Diseases Laboratory. The laboratory needs to conduct transcriptome and immune profiling analyses on 32 tissue samples from cattle, small ruminants, and lab animals. The reagent kits must enable single cell transcriptional and immune profiling of millions of cells extracted from various tissue types. They should allow single cell suspensions to be partitioned into thousands or millions of Gel Beads-in-Emulsion with unique barcodes and be compatible with the existing Chromium Controller for precise reagent delivery to over 80,000 micro-reactions in parallel. The kits must include reagents, unique dual index oligonucleotides, and a minimum of four microfluidics chips to handle all 32 samples representing a wide range of eukaryotic cell sizes and types. They should also provide unlimited free access to bioinformatics and visualization software tools for end users of varied experience levels. The performance period is from September 15, 2023 to November 30, 2023.
The related federal contract opportunity is solicitation number 12305b23q0132 for "Reagent Kits" being procured by the USDA Agricultural Research Service Field Research Implementation and Information Delivery Northeast Area.

Statement of work
Background
USDA-ARS Animal Parasitic Diseases Laboratory (APDL), in Beltsville, Maryland is conducting scientific research to understand host-parasite interactions using advanced single cell genomics technologies. APDL scientists have 32 samples collected from gut tissues of cattle, small ruminants, and lab animals and need to conduct transcriptome and immune profiling analyses at a single cell resolution from complex tissues. The success of single cell genomics relies on high quality and specific reagent kits for the library construction and downstream analyses.
Requirements
The detailed specification of reagent kits includes:
1. The reagent kits shall enable single cell transcriptional and immune profiling of up to millions of single cells extracted from 32 different tissue sample types.
2. The kits should allow single cell suspensions to be partitioned into thousands or millions of Gel Beads-in-Emulsion (GEMs) with unique barcodes for downstream applications.
3. The assay formulation should be flexible, compatible with the low throughput, standard and high throughput (96-well plate) workflows, with high cell capture efficiency up to 60%.
4. The reagent kits should be compatible for the existing Chromium Controller, for a precisely engineered reagent delivery method that enables > 80,000 of micro-reactions in parallel.
5. The kits should include both reagents and unique dual index oligonucleotides in a 96-well plate form.
6. The kits should include minimal four microfluidics chips, capable of handling 32 tissue samples, with a wide range of eukaryotic cell sizes and cell types.
7. The kits should be versatile with the applications in tagging single cells, individual nuclei, guide RNA, cell surface proteins labeled with antibody-oligonucleotide conjugates, or CRISPR transduced cells.
8. The purchase of reagents should include free and unlimited access to bioinformatic pipelines and visualization software tools for end users, both novice and expert alike.
9. The reagent kits should be delivered in two (or more) batches, per scientists’ requests.
Period of Performance (POP) for the requisition From Sept 15th, 2023 to Nov 30th, 2023.
