STATEMENT OF REQUIREMENTS
Droplet Generator
I. BACKGROUND
The Biomarker and Genomic Sciences Group within the Materials Measurement lab (MML) at
(NIST) needs to acquire a droplet generator for droplet digital polymerase chain reaction
(ddPCR) to make absolute quantifications of Standard Reference Materials (SRM) using the Bio-
Rad QX200 ddPCR instrument NIST currently owns. This will allow a higher throughput of samples and will allow workflow to continue if our current droplet generator must be serviced.
II. SPECIFICATIONS
The Contractor shall provide one (1) droplet generator that meets the following specifications:
• Droplet generator must be compatible with existing Bio-Rad QX200 Droplet Digital PCR
Reader, meaning positive and negative droplets can be read using the QX200 droplet reader and is compatible with either QuantaSoft or QX Manager software.
• Droplet generator must be able to use Bio-Rad Supermix for Probes without 2´-
Deoxyuridine, 5´-Triphosphate (dUTP), catalog numbers 1863023, 1863024, 1863025.
• Droplet generator must be able to use the DG8 cartridges for droplet generation which holds a total of eight reactions.
• Droplet generation must use oil to generate droplets in conjunction with the Bio-Rad
Supermix for Probes (no dUTP)
• Droplet generator must be able to partition a 20 microliters (µL) reaction into approximately 20,000 nanoliter-sized droplets for up to eight samples at a time.
• Droplet volume and size must be consistent between runs (approximately 1 nanoliter (nL) volume and a diameter of approximately 120 micrometers (µm).
• Droplets must be generated in approximately 2 minutes for 8 samples.
OPTION FOR ADDITIONAL PURCHASE
The contractor shall provide one (1) additional droplet generator, to be exercised at the discretion of NIST within one year after contract award. Delivery shall occur within 45 calendar days of exercise of option.
III. DELIVERY
Delivery shall be FOB Destination and shall occur within 45 calendar days of award. Delivery shall take place during normal business hours, between 8:30 am and 5:00 pm Eastern Time, Monday through Friday except Federal Holidays, and shall be coordinated with the NIST
Technical Point of Contact (TPOC). FOB Destination means: The contractor will pack and mark the shipment in conformance with carrier requirements, deliver the shipment in good order and condition to the point of delivery specified in the purchase order, be responsible for any loss of and/or damage to the goods occurring before receipt and acceptance of the shipment by the consignee at the delivery point specified in the purchase order; and pay all charges to the specified point of delivery. Delivery shall be made to the NIST campus at 100 Bureau Drive, Gaithersburg, MD, 20899, USA.
IV. INSTALLATION
Not required.
V. TRAINING
Not required.
VI. WARRANTY
The Contractor shall provide, at minimum, a one (1) year warranty to include all parts, materials, labor, and travel for the instrument to keep the instrument operating to manufacturer's specifications. The warranty period shall commence upon delivery.
VII. ACCEPTANCE
Acceptance shall occur when the instrument is tested by the NIST TPOC within 7 calendar days of delivery. TPOC will test a no template control and the SRM 2373 (HER2 copy number) material to determine if the instrument is working appropriately based on whether the results are within the range of NIST’s certified values for SRM 2373.
VIII. DELIVERABLES
Description Quantity Due Date
