Optical Coherence Microscope (OCM)
I. BACKGROUND
An optical coherence microscope (OCM) is required to make non-invasive, label-free, 3D measurements of cell viability in large fields of view within tissue engineered hydrogels. The hydrogel scaffolds make it difficult to image cells to assess their viability when the cells are contained with the hydrogels, which is possible with the OCM.
II. SPECIFICATIONS
[1] OCM will be a spectral domain OCM (SD-OCM).
[2] OCM will use long wavelength 930 nm light with an optical bandwidth of > 100 nm to enhance tissue penetration.
[3] Imaging depth will be 2.7 mm in air and 2.0 mm in water.
[4] Axial Resolution will be 6.0 μm in air and 4.5 μm in water.
[5] Lateral Resolution will be 8 μm.
[6] Light source will be a superluminescent diode (SLD).
[7] OCM will include a large field of view scanning lens with a lateral resolution of 8 micrometers, a working distance of 25 mm, a 10 mm X 10 mm field of view.
[8] OCM will include a second scanning lens with a smaller field of view with a lateral resolution of 4 micrometers, a working distance of 7.5 mm and a 6 mm X 6 mm field of view.
[9] OCM maximum sensitivity will be 101 dB at 5 kHz.
[10] OCM will have ability to detect motion by either Doppler mode or by speckle variance model.
[11] OCM shall have a video camera and light to aid in the position of samples and to document the region of the sample that is being imaged by OCM.
[12] OCM will come with a stand to mount the scanning lens with coarse and fine focus knobs.
[13] OCM shall allow user flexibility in controlling image dimensions and sampling density (voxel size).
[14] Pixel dimensions across a 2D image shall be adjustable between 16 pixels to 10000 pixels.
[15] OCM will have adjustable A‐scan/line rate of 5 kHz to 248 kHz for high sensitivity or high-speed scanning.
[16] OCM will have an automated stage for automated collection of data.
a. Travel range of 110 mm x 75 mm
b. Maximum velocity of 250 mm/s
c. Maximum acceleration of 2000 mm/s^2
d. Bidirectional repeatability of 0.25 µm
e. Unidirectional repeatability of 0.25 µm
f. Horizontal load capacity of 1.0 kg
g. Minimum achievable incremental movement of 0.1 µm
h. Home location accuracy of 0.25 µm
i. Absolute on-axis accuracy of <3 µm
j. Joystick for ease of manipulation [17] OCM system will include a computer, software, monitor, mouse and keyboard to run the microscope.
[18] OCM will include intuitive software so that a non-technical expert can run the unit.
[19] Reference arm will be integrated in probe head (common path) to prevent influence of polarization by moving fiber for all scanner types.
[20] OCM shall allow export of images in several file types including tiff, jpg and pdf.
[21] Collected data shall be accessible in both raw and processed formats to enable the user to develop novel analytical routines.
[22] The computer that runs the OCM must be compatible with NIST network (TPM 2.0 compatible). This can be achieved two ways. First, a NIST computer can be used to run the OCM if the proper ports and software can be installed on a NIST Dell Win 10 machine. The second option is to provide a NIST compatible computer, which must be a Dell from the following list:
a. Dell Latitude Laptops & Tablets that support TPM 2.0: 3180, 3189, 3380, 3390, 3470, 3480, 3488, 3570, 3580, 3588, 5175, 5179, 5280, 5285, 5288, 5289, 5290, 5414, 5480, 5488, 5490, 5580, 5590, 5591, 7200, 7202, 7214, 7275, 7280, 7290, 7300, 7370, 7380, 7389, 7390, 7400, 7414, 7470, 7480, 7490, E5270, E5370, E5470, E5570, E7270, E7370 and E7470.

III. DELIVERY
The Contractor shall deliver the instrument within 90 days of the award. However, due to current access restrictions, delivery shall not take place until the Contractor coordinates with the designated TPOC and Contracting Officer (CO). Prior to shipment, the Contractor must ensure NISTs campus is open and accepting deliveries. IF the restrictions of access to the campus impacts delivery schedule, a modification may be executed by the CO to establish an extended delivery timeframe.
IV. INSTALLATION
The contractor shall install the instrument within 90 days of the award. Installation shall include, at a minimum, uncrating/unpacking of all equipment, setting up the computer and software to operate the OCM, start-up, demonstration of specifications, and removal of all trash. The Instrument will operate to manufacturer’s specifications upon installation. The contractor must demonstrate that the OCM is working according to manufacturer’s specifications and must use test specimens.
V. TRAINING
The Contractor shall provide training for three NIST personnel on the NIST Campus, Gaithersburg, MD within 90 days. Training shall include OCM operation, maintenance, troubleshooting and use of software.
VI. WARRANTY
The contractor shall provide, at minimum, a one-year warranty on the OCM. The warranty shall include all parts, computer, software, labor and travel. Warranty shall commence upon successful completion of delivery, installation, training, and demonstration of all required specifications.
