SPECIFICATION DOCUMENT
BACKGROUND
USDA, Agricultural Research Service (ARS), Animal Parasitic Diseases Laboratory (APDL), is conducting scientific research to understand the evolution and host-parasite interaction using population and comparative genomics. APDL scientists are routinely conducting whole genome sequencing of eukaryotic protozoan pathogens to understand the genome evolution, population genetic structure, zoonotic transmission, and outbreak potential of these pathogens. The success of whole genome sequencing relies on high quality and specific and accurate size selection of DNA fragments for the library construction in Next Generation Sequencing (NGS) workflows.
REQUIREMENT
DNA Size Selection System, brand name or equal to a BluePippin Size Selection that allows us to conduct DNA size selection using a pulsed-field electrophoresis necessary to conduct our next generation sequencing research.

INSTRUMENT CAPABILITIES
Must be capable of separating DNA by agarose gel electrophoresis in disposable pre-cast gel cassettes. After size selection, we must be able to electro-elute DNA (50 bp to 50,000 bp) from agarose-filled sample lanes into the electrophoresis buffer.
Gel electrophoresis must be capable of running direct current electrophoresis and pulsed-filed electrophoresis.
Individual sample lanes must be separated by a physical barrier.
Each sample lane must be bifurcating and be electrophoretically controlled by switchable positive electrodes for accurate fragment size selection, which should be regulated by a computer software system.
Must have multiple lanes capacity to simultaneously purify several samples.
Accurate size selection can be determined by the migration rate of the DNA size ladder, which will be used as an internal or external standard.
