Statement of Need Fully automated vibrating blade microtome

1. Background Vibrating blade microtomes are used to produce monolayer or thick sections for fixed or fresh tissue under physiological conditions without freezing or embedding. Sectioning fresh tissue specimens with Leica VT Series microtome maintains the morphology, enzyme activity and cell viability of the tissue while also minimizing artifacts, compression distortion, cell destruction and other inherent deleterious effects of sectioning

2. Objectives The vibrating blade microtome features a motorized blade holder with a sectioning speed adjustable between 0.01 to 1.5 mm/second. The blade holder can be rotated through 90o to permit accurate insertion of a whole double-edged razor blade. Built-in LED illumination provides natural, comfortable lighting during sectioning without adding heat, which could deteriorate the tissue. The Leica VT1200 S features 5-step adjustable light intensity.

3. Scope The fully automated vibrating microtome can be used in both semi- or fully automated sectioning modes and offers automatic feeding, specimen retraction, and a cutting window of 0.5mm to 45 mm. Section thickness adjustment in 1 um increments up to a maximum of 1000 um. The mode of operation can be individually selected, and settings can be stored or up to 8 users. Automatic feeding, specimen retraction and use of a cutting window are designed to minimize sectioning time, without the need of replicating experiments.

Applicable Directives The fully automated vibrating blade microtome with Vibrocheck includes double-walled buffer trays in two different sizes which allows sectioning of specimens 33.x 40 mm or as large as 70 x 40 mm, chiller cooled. Fully automated cut mode, blade travel speed of 0.01-1.5 mm/second. During operation, the blade vibrates laterally and advances forward through the specimen.

4. Performance Requirements

The highly specialized device fulfills the following specifications:

Fully automated vibrating blade microtome for sectioning unfixed specimen from Neuropathology and Neurophysiology (Patch clamping).

Built-in software remembers individual operational parameter settings for up to 8 users.

5. Deliverables

b. Installation of microtome in Robinson Lab space, CSB Room 309.

c. Warranty, to include (1) year of full coverage on parts, labor, and software, to include: remote diagnostic repair support, all replacement parts, travel time and labor for onsite assistance, software updates and safety updates and (1) preventative maintenance visit in year two.
