PERFORMANCE WORK STATEMENT

GENERAL INFORMATION

A. Introduction

The USGS Upper Midwest Water Science Center Michigan Bacteriological Research Laboratory (MI- BaRL) has project work that aims to understand the influence of resuspended cyanobacteria cells from the sediment on seeding algal blooms in water column. For this work, water and sediment samples are collected in incubated in selective cyanobacteria growth media. Cultivated cyanobacteria growth is then identified to determine “who’s growing” in the sediment and water column and whether similar species found in the sediment are also found in algal blooms. Subsequently, this work requires expert microscopic identification of preserved algal cells in our cultures.

B. Background

USGS scientists are collaborating with Federal, State, and local agencies to understand environmental influences of toxin-producing cyanobacteria in aquatic environments used for drinking water or recreational purposes. This project aims to make the connection between resuspended sediment and lake water column cyanobacteria communities by (1) assessing cyanobacteria growth and community indicators spatially and temporally and 2) comparing the timing of nutrient flux and sediment resuspension to the water column with algal bloom initiation.

C. Scope

Collaboration for analysis and reporting are needed for this study. Samples require identification and reporting of algal species present in the preserved culture growth. Due to variable nature of algal blooms and need to identify algal bloom species rapidly to respond to collaborator and cooperator needs, we require that the contractor has the capabilities and offers services including providing permanent archival slides, digital images, microcystin testing, and semi-automated IFCB analysis, and enumeration of phytoplankton.

WORK REQUIREMENTS

A. Technical Requirements

The Contactor shall:

1. Provide instructions for sample preservation and shipment.

2. Provide analyst request forms and electronic submission of forms.

3. Confirm receipt of samples.

4. Have experience working with samples preserved using glutaraldehyde.

5. Be able to receive and process samples preserved using glutaraldehyde.

6. Identify phytoplankton to division, genus, and species level.

7. Upload sample information, processing status, and results into an electronic data management system.

8. Collaborate on final manuscript by providing analytical methods description and input on interpretation of results.

PAGE OF PAGES

9. Upon request, be able to create optically clear, permanent archival slides and safely store.

10. Upon request, be able to capture and provide high-resolution images of sample specimen.

B. Deliverables

The contractor shall:

1. Provide data summaries on phyla, division, class, genus, and species present in samples.

2. Provide a summary of analytical methods and interpretation of data.

3. Provide analysis on diversity indices and summary statistics.

4. Report results in a machine-readable format.

SUPPORTING INFORMATION

A. Period of Performance:

Base: August 01, 2022 – July 31, 2023 Option Year One: August 01, 2023 – July 31, 2024 Option Year Two: August 01, 2024 – July 31, 2025 Option Year Three: August 01, 2025 – July 31, 2026 Option Year Four: August 01, 2026 – July 31, 2027

The contractor shall deliver data no later than 150 days following receipt of samples.

The contractor shall archive samples in -80 freezers The contractor shall perform re-runs if there are questions regarding data quality The contractor shall ship samples to collaborators upon request The contractor shall provide contributions to the manuscript within 240 days following receipt of samples.
