STATEMENT OF WORK
TITLE: Procurement of a Plate reader LAB REQUESTING SERVICE: Material Measurement Laboratory, Biomolecular Measurement Division
I. BACKGROUND INFORMATION
The Macromolecular Structure and Function (MSF) Group in the Biochemical Science Division of the Materials Measurement Laboratory at the National Institute of Standards and Technology (NIST) advances novel measurement methods and models for the determination and analysis of the structure and function of biological macromolecules, including lipids, DNA, RNA, proteins, and macromolecular complexes and assemblies formed by these molecules; advancing methods to determine the physical and chemical factors which determine the specific interactions of macromolecules with cofactors and ligands; studies the relationships between biomolecule structure, dynamics, function, and energetics using computational, biophysical, and biochemical techniques; develops reference databases for these properties; and advances measurement methods for engineering complex biological systems.
Recently, the MSF group received a NIST Innovations in Measurement Science (IMS) Program on Next Generation Protein Sequencing. To achieve the goals of the IMS program the group needs a plate reader that can be used for several experiments including monitoring cell growth and protein expression, enzyme-linked immunosorbent assay (ELISA) and Fluorescence polarization anisotropy (FPA).
II. SPECIFICATIONS
The Contractor shall provide one (1) Plate reader that meets the following minimum technical requirements. The Contractor shall install the instrument and provide training as described below.
System Requirements:
The Multi-mode plate reader shall meet the following minimum requirements:
· The plate reader shall be new and not refurbished.
· The plate reader shall have absorbance, fluorescence, and luminescence modes:
· The absorbance mode wavelengths shall be from at least 230-950nm and have a photometric range of 0-4.0 OD.
· The fluorescence mode shall be hybrid such that it is both monochromator (250-850nm range in 1nm increments and a variable bandpass filter selection in < 5nm increments) and filter based.
· Top fluorescence sensitivity shall be <1pM (96 well plate), and bottom fluorescence sensitivity shall be <5pM (96 well plate).
· The luminescence sensitivity shall be <1femtomole (ATP) with a 6-decade dynamic range.
· The instrument shall have separate optics for bottom reads (filter based).
· The filters must be suitable for fluorescence polarization with a sensitivity <5mP at 1 nM fluorescein.
· The plate reader shall control temperature in range of at least 25 to 55°C.
· The instrument shall be able to accommodate 6-384 well plates, microvolume cuvettes, and 2 μL microspots (with appropriate attachments) and adjust pathlength based on volume.
· The instrument shall have a dual reagent injector capable of dispensing volumes in 1 μL increments.
· The instrument shall come with at least 3 licenses of the controlling software to run the instrument and analyze the data.
· The instrument shall come with filters to perform fluorescein (~494ex/512em), and TAMRA (~546ex/579em) based assays and fluorescein-based fluorescence polarization assays.
· The instrument shall have the ability to be upgraded to hook up to a gas controller.
Installation:
The Contractor shall install the instrument in Building 1A, Room 218, at the National Institute of Standards and Technology (NIST)/Institute for Bioscience and Biotechnology Research (IBBR) located in Gaithersburg, MD. Installation shall include, at a minimum, uncrating/unpacking of all equipment, set-up and hook-up of all equipment, start-up, demonstration of specifications, and removal of all trash. Instrument must operate to manufacturer’s specifications upon installation. The manufacturer’s specifications (required upon quote submission) will be incorporated into the resultant purchase order. Installation shall take place during normal business hours, between 8:30 am and 5:00 pm Eastern Time, Monday through Friday except Federal Holidays, and shall be coordinated with the NIST Technical Point of Contact (TPOC). Installation must be within 60 days from award date.
Warranty:
The Contractor shall provide, at a minimum, a one-year warranty for the equipment. The warranty shall cover all parts, labor and travel. The warranty shall commence upon successful completion of delivery, installation, training and demonstration of all required specifications.
Training:
Include training at NIST/IBBR for up to 4 NIST/IBBR personnel covering normal operation, troubleshooting, and routine maintenance. Training will be provided during normal business hours, between 8:30am and 5:00pm Eastern Time, Monday through Friday, except Federal Holidays, and will be coordinated with the NIST Technical Point of Contact (TPOC) to ensure maximum availability of NIST personnel. Training must occur within 10 days of installation and may take more than one day.
