STATEMENT OF WORK

TITLE: Procurement of two liquid chromatography systems LAB REQUESTING SERVICE: Material Measurement Laboratory, Chemical Sciences Division

I. BACKGROUND INFORMATION

The National Institute of Standards and Technology (NIST), Organic Chemical Measurement Science group within the Chemical Sciences Division is dedicated to organic chemical measurements in the areas of clinical, environmental, nutritional, and forensic chemistry. A significant portion of this work involves quantitative measurements of organic compounds in standard reference materials (SRMs). This effort includes the use of liquid chromatography (LC) instrumentation paired with spectroscopic (ultraviolet/visible absorbance (UV/Vis) and fluorescence (FL)) detection systems. Also, the LC systems can be paired with mass spectrometers (MS) to provide additional detection methods. An LC procured for these purposes must enable the separation of many small organic molecules in complex matrices for detection by UV/Vis, FL, and/or MS systems.

Currently, there are operating LCs that are in use with UV/Vis, FL, and MS detection systems, although their performance specifications do not allow the LCs to operate at liquid pump pressures above 400 bar. Due to this limitation, many of the chromatographic separation methods developed in the Organic Chemical Measurement Science group cannot use liquid chromatography columns with smaller inner diameters and/or small particle sizes of the stationary phase silica. Methods using these aspects of columns would provide higher-throughput and, potentially, better chromatographic quality.

NIST requires two complete LCs that are compatible with current MS instrumentation and software currently in use, specifically an Agilent 6490 triple quadrupole MS with Mass Hunter software and AB Sciex 4000 triple quadrupole MS with Analyst software. Both LCs must communicate with and be controlled by a single point interface with the MS software.

II. SCOPE OF WORK

The scope of the this work is to procure two complete LCs capable of operating at liquid pump pressures of 600 bar or better, which would include UV/Vis and FL detection systems.

III. SPECIFICATIONS

The Contractor shall provide two (2) analytical instrument systems that meet the respective minimum technical requirements. The Contractor shall install the instrument and provide training as described below.

System Requirements:

Liquid Chromatograph #1 (LC-1)

· Shall contain the following modules: solvent storage tray, mobile-phase degasser (can be combined with solvent tray or solvent pump), binary solvent pump (with solvent selection valves for at least four solvents), temperature-controlled autosampler, temperate-controlled column heater/cooler with 6-port switching valve, photodiode array UV/Vis absorbance detector, and fluorescence detector. All modules must be controlled by a single software interface, as described below.

· Shall have a single point interface with AB Sciex Analyst software and Agilent MassHunter software, i.e. the LC instrument must be completely controlled by both of these software packages via communication cables.

The instrument shall meet the following module-specific requirements:

Solvent Pump w/ degasser

· Flow rate range of 0.05 – 5 mL/min

· Flow precision of ≤ 0.07% RSD or 0.01 min SD (whichever is greater)

· Flow accuracy of ± 1% or ± 10 µL/min (whichever is greater)

· Maximum operating pressure of 600 bar or greater

· Solvent selection valve for up to four solvents Autosampler with temperature control

· Maximum operating pressure of 600 bar or greater

· Injection range of 0.1 to ≥ 100 µL

· Injection accuracy of ≤ 1 %

· Carryover of less than 0.004 %

· Includes vial tray for standard 2 mL autosampler vials

· Temperature range of 4 – 40 ºC

· Temperature accuracy of ± 2 ºC

· Communicates via controller-area network (CAN) Temperature-controlled column compartment

· Temperate control range from 10 ºC below ambient to 80 ºC

· Temperature stability of ± 0.15 ºC

· Temperature accuracy of ± 0.5 ºC

· Include a 6-port/2-position valve, controllable by LC software Photodiode Array UV/Vis Detector

· 1024-element diode array detector with deuterium light source

· Wavelength range of 190 – 640 nm

· Maximum sampling rate of 80 Hz

· Wavelength accuracy of ± 1 nm

· Included flow cell with maximum operating pressure of at least 70 bar.

Fluorescence Detector

· Maximum data scan rate of 74 Hz or greater

· Excitation wavelengths by xenon lamp of 200 to 1200 nm

· Emission wavelength detection of 200 to 1200 nm

· Included flow cell with maximum operating pressure of at least 20 bar

· Detection of up to 4 emission wavelengths in a single analysis

Liquid Chromatograph #2 (LC-2)

· Must contain the following modules: solvent storage tray, mobile-phase degasser (can be combined with solvent tray or solvent pump), quaternary solvent pump, temperature-controlled autosampler, temperature-controlled column heater/cooler with 6-port switching valve, and photodiode array UV/Vis absorbance detector. All modules must be controlled by a single software interface, as described below.

· Shall have a single point interface with AB Sciex Analyst software and Agilent MassHunter software, i.e. the LC instrument must be completely controlled by both of these software packages via communication cables.

The instrument shall meet the following module-specific requirements:

Solvent Pump w/ degasser

· Flow rate range from 0.001 to 5 mL/min

· Flow precision of ≤ 0.07% RSD or 0.01 min SD (whichever is greater)

· Flow accuracy of ± 1% or ± 10 µL/min (whichever is greater)

· Pressure range up to 1300 bar at 0 – 2 mL/min flow

· Active seal wash capabilities Autosampler with temperature control

· Pressure range up to 1300 bar

· Injection range of 0 to ≥ 20 µL

· Carryover of less than 0.0009 %

· Includes vial tray for standard 2 mL autosampler vials

· Temperature range of 4 – 40 ºC

· Temperature accuracy of ± 2 ºC Temperature-controlled column compartment

· Temperature range of 20 ºC below ambient to 110 ºC

· Temperature stability of ± 0.05 ºC

· Temperature accuracy of ± 0.5 ºC

· Include a 6-port/2-position valve, controllable by LC software Photodiode Array UV/Vis Detector

· 1024-element diode array detector with deuterium light source

· Wavelength range of 190 – 640 nm

· Maximum sampling rate of 80 Hz

· Wavelength accuracy of ± 1 nm

· Included flow cell with maximum operating pressure of at least 70 bar.

Installation:

The Contractor shall install the instrument in Building 227, Room B163 and B159, at NIST Gaithersburg, MD. Installation shall include, at a minimum, uncrating/unpacking of all equipment, set-up and hook-up of all equipment, start-up, demonstration of specifications, and removal of all trash. The instrument shall operate to manufacturer’s specifications upon installation. Installation will take place during normal business hours, between 8:30 am and 5:00 pm Eastern Time, Monday through Friday except Federal Holidays, and will be coordinated with the NIST Technical Point of Contact (TPOC). Installation shall occur within 90 days of award.

Warranty:

The Contractor shall provide, at a minimum, a one year warranty for the equipment, which includes maintenance services, including scheduled/preventative maintenance. The warranty shall cover all parts, labor and travel. The warranty shall commence upon successful completion of delivery, installation, training and demonstration of all required specifications.

Training:

The Contractor shall provide training at NIST for up to 2 NIST personnel covering normal operation, troubleshooting, and routine maintenance. Training shall be provided during normal business hours, between 8:30am and 5:00pm Eastern Time, Monday through Friday, except Federal Holidays, and will be coordinated with the NIST Technical Point of Contact (TPOC) to ensure maximum availability of NIST personnel. Training shall occur within 10 days of installation.

IV. PERIOD OF PERFORMANCE

The period of performance shall be one (1) year from the date of acceptance.

V. PLACE OF PERFORMANCE

All work shall be completed at the Contractor’s facility. Installation of the instrument and training shall be accomplished at NIST, Gaithersburg, Maryland. Normal duty hours are Monday through Friday, 8:30 a.m. to 5:00 p.m. with the exception of Federal holidays.

VII. ACCEPTANCE

Acceptance will occur after the Contractor demonstrates that all performance standards in this statement of work have been met, including training of NIST personnel.

VIII. GENERAL INFORMATION

Safety: The Contractor employee shall be responsible for knowing and complying with NIST installation safety prevention regulations (http://www-i.nist.gov/mml/safety/policies/index.htm.) Such regulations include, but are not limited to, general safety, fire prevention, and waste disposal.

Patent Rights: The Government retains a Government use license to all inventions arising from this work.

Security: NIST is a restricted campus. An identification badge is required for access for entry into buildings and also is shown to the armed Security Police when entering the campus.

Identification Badges: Contractor employees shall comply with NIST identification and access requirements. The Contractor employee is responsible for absences due to expired identification and access documents. Each Contractor employee shall wear a visible identification badge provided by the NIST Security Office. The badge must show the full name, title, and if required by NIST, the words “Contractor” in front. The Contractor employee shall turn in the NIST identification badge and vehicle pass to the TPOC, COR, or Contracting Officer (CO) upon termination of their services under this contract.

Vehicle Registration: All Contractor employees must register their vehicles with the NIST Security Office to gain access to the campus. A valid driver’s license, Government-furnished civilian ID, proof of insurance and current registration must be presented to the NIST Security Office, at which time a NIST vehicle pass will be issued. The pass shall be displayed on the vehicle’s rear view mirror in accordance with instructions. The Contractor employee shall follow NIST procedures for removal and turn-in of the vehicle pass upon termination of services under this contract.
