Statement of Work (SOW) Cryogenic Scanning Electron-Raman Microscope System

FDA Project No: CDER-2024-121914

1. Scope of Work To purchase and install a field emission gun (FEG) scanning electron microscope (SEM) integrated with a Raman microscope and capable of analyzing cryogenically fixed samples to expand and augment the capacity and capability of Center for Drug Evaluation and Research, Office of Pharmaceutical Quality (CDER/OPQ) to ensure quality, safety, and efficacy of pharmaceutical products. In particular, this system will offer highest spatial resolution and multi-attribute characterization of complex and other drug products, thus promote use of advanced technologies in domestic manufacturing by implementing advanced analytical methods to evaluate products.

2. Statement of Need

a) Background Scanning electron microscopy (SEM) is an essential and well-established tool used for image-based characterization of most pharmaceutical products but especially products containing particles such as powders, granules, colloidal suspensions at different length scales as well as solid forms. Modern or state-of-the-art SEMs provide sub nanometer resolution with the use of a field emission gun (FEG) and sometimes are referred to as FESEM.

Electron microscopy generally requires sophisticated sample preparation due to high vacuum environment required to monitor the interaction between the electron beam and the specimen. Hence, pharmaceutical formulations that are colloidal and/or in aqueous or other liquid environments need to be chemically or cryogenically fixed for analysis in a native-like state. Tools that are generally required for cryogenic SEM (cryo-SEM) include cryotransfer loading system and shuttle, freeze fracture, freeze-coating, cryogenic ion beam milling and temperature controlled cryo-stage.

Interaction of an electron beam with a specimen produces various types of electrons such as backscattered, secondary, Auger as well as characteristic and continuum X-rays. Therefore, specialized detectors may be required to register images of samples with topographical and compositional information. Electron detectors also vary on operation and sensitivity at high or low voltages or vacuum. In addition, X-ray detectors are utilized in SEM to provide elemental mapping via characteristic X-rays. This technique is called energy dispersive X-ray spectroscopy (EDS).

In advanced SEM systems, Raman spectroscopy can be utilized as an additional chemical detection system to provide molecular identification and multivariate chemical imaging, referred to as SEM-Raman interface systems.

An integrated cryo-SEM-Raman system with a FEG is an advanced analytical tool capable of providing elemental, chemical, and physical/topographical characterization of a pharmaceutical product with highest spatial resolution. Use of advanced technologies in domestic manufacturing of drug products utilizes tools like cryo-SEM-Raman in various stages of product development as well as quality control and finished product testing. To assess implementation of SEM-related methods and techniques developed by industry in domestic manufacturing and to characterize complex or otherwise pharmaceutical products using the cryo-SEM-Raman advanced analytical platform, the agency may acquire and research an FEG cryo-SEM- Raman system that can also retrofit a present EDS detector.
