About this file
This document is a Statement of Requirement for the procurement of a precision two-axes translation stage and controller for fluorescence microscopy. The National Institute of Standards and Technology (NIST) requires this equipment to support their research in fluorescence correlation spectroscopy and fluorescence microscopy for characterizing biopharmaceutical solutions and materials. The key technical requirements include: a two-axis translation stage with 25 mm x 25 mm travel range, 100 mm/s maximum velocity, 0.1 μm positioning resolution, and a 76 mm x 76 mm clear aperture. The motion controller must support multiple communication interfaces and programming APIs. The contractor shall provide one unit of the specified equipment and an operation manual within 160 days of award. The government will evaluate quotations based on technical acceptability and lowest price.

STATEMENT OF REQUIREMENT
TITLE: Procurement of a precision two-axes translation stage and controller for fluorescence microscopy
I. BACKGROUND INFORMATION
The Materials Science and Engineering Division (MSED) is involved in the advancement of fluorescence correlation spectroscopy with imaging capabilities for the characterization of biopharmaceutical vaccine solutions, networks, and gels. These materials often form structures on the 100 micron length scale due to instability, aggregation, or phase separation. This process leads a non-uniform distribution of components, such as polymer, RNA, protein, surfactant, and solvents. The spatial positions of such components that are fluorescently labeled can be tracked by fluorescence microscopy (FM), while the diffusion characteristics by fluorescence correlation spectroscopy (FCS). In order to combine these capabilities a focused laser spot must be precisely collocated at predefined positions with sub-micron spatial resolution in the fluorescence field of view.
II. SPECIFICATIONS
The Contractor shall provide one (1) XY positioning stage and associated motion controller that meets the following minimum technical requirements. The two-axes stage must fit within the frame of our existing apparatus and have a clear aperture to permit epi-illumination or trans-illumination. The two-axes stage will require automated broad range of translation, precision translation resolution, encoding of positions, low noise and drift achieved by piezo-actuation, and fast motion velocity.
System Requirements:
Two axes translation stage
• Surface mountable upon an existing platform where two-axes maximum travel range fit within of 190 mm x 190 mm platform.
• Compact vertical dimensions of less than 40 mm to fit within existing 200 mm space.
• Independent (X × Y) travel range of at least 25 mm × 25 mm
• Maximum velocity of 100 mm/s in X axis
• Maximum velocity of 100 mm/s in Y axis
• Encoded axes to allow for repeatable repositioning by software.
• 0.10 micrometer positioning resolution or better
• Repeatable positioning of +/- 0.3 micrometers or better
• 0.10 micrometer or better incremental position resolution
• Noncontact limit to prevent movement beyond defined limit.
• A minimum of 76 mm x 76 mm clear aperture for placement of a standard 76 mm × 25.4 mm (3” × 1'') microscope slide directly upon or via insertable holder.
• Less or equal to +/- 50 microradian angular rotational motion (pitch and yaw) along X, Y, and Z directions.
• XY stage should be less than 2 kg total mass as required by placement upon an existing z-scan stage.
Motion Controller
• Shall control a minimum of two independent axes.
• USB, RS-232, TCP/IP, SPI communication interfaces shall be included.
• Shall support application programming interfaces including Python and MATLAB and drivers for
National Instruments LabView
• Macro programmable for stand-alone functionality.
• Shall be capable of programmable motion profiles.
• Operation with analog or digital joystick for motion profiles and positioning.
• An operation manual either electronic or hard copy shall be included.
Warranty:
The Contractor shall provide, at a minimum, a one-year warranty for the equipment. The warranty shall cover all parts, labor and travel. The warranty shall commence upon Government acceptance.
III. DELIVERABLES
