Version 11-30-2016-1 FAR Part 11—Describing Agency Needs
PURCHASE DESCRIPTION
1. Statement of Need and Purpose:
The National Institute of Mental Health (NIMH), Section on Critical Brain Dynamics currently has a Bergamo II 2-Photon Imaging (2-PI) microscope for experimental research, which was purchased approximately 5 years ago. The technology underlying 2-PI since then has advanced to include more flexible scanning capabilities. The current requirement focuses on the upgrade of the existing microscope as well as replacement of degraded modules to significantly enhance and stabilize the process of the 2-PI. This upgrade is necessary to continue to produce experimental data relevant for understanding the role of brain circuits in normal and pathological behavior.
2. Background Information and Objective:
The Brain Research Through Advancing Innovative Neurotechnologies (BRAIN) initiative, is a US-wide collaboration of brain scientists and engineers to explore new frontiers in recording from brains at high temporal and spatial resolution. By accelerating the development and application of innovative technologies, researchers will be able to produce a revolutionary new dynamic picture of the brain that, for the first time, shows how individual cells and complex neural circuits interact in both time and space. The 2-PI microscope was specifically combined with a second, experimental platform to develop spatial-light-modulated (SLM) laser controls. The SLM-stimulation path is mounted on top of the original microscope and allows precise optogenetical stimulation of single nerve cells deep inside the cortex of wake animals during sensory stimulation and animal behavioral outcome. Both, 2-PI and SLM platforms are custom-designed and comprise one of the few all-optical interrogation set-ups worldwide. The unique nature of this setting, its close reliance on Thorlabs specific modules with respect to the optical and mechanoptical controls require regular custom-updates and custom-replacements of specific modules. This all-optical, experimental platform continues to be a part of an ongoing U19 BRAIN initiative grant that includes a world-wide collaboration of eleven
(11) groups.
