About this file

This sources sought notice requests capability statements from small businesses for the purchase of long read sequencing kits by the National Institute on Aging, Center for Alzheimer's Disease and Related Dementias. Interested vendors must be able to provide kits that deliver paired full-length T-cell receptor alpha and beta chain sequences at the single cell level using universal amplification strategies to avoid bias, and incorporate unique molecular identifiers to further reduce bias and enable transcript quantification. Responses are requested to identify how vendors' proposed products meet requirements, provide literature demonstrating ability to supply within one month of a completed purchase order, and state business size and type. Responses must be received by January 12, 2022. The notice seeks to define the procurement strategy for a forthcoming solicitation.

Version 11-30-2016-1 FAR Part 11—Describing Agency Needs

PURCHASE DESCRIPTION

1. Statement of Need and Purpose: This acquisition is necessary to provide supplies and reagents to the Center for Alzheimer’s Disease and Related Dementias (CARD) for neurodegenerative disease research for gene expression analysis.

2. Background Information and Objective: CARD will create a national resource of isogenic sets of Alzheimer’s Disease and Related Dementias mutation iPSC lines; complimented by foundational phenotypic datasets from differentiated disease-relevant cells derived from these lines. CARD anticipates that this resource will substantially accelerate the pace of mechanistic discovery regarding how Alzheimer’s Disease and Related Dementia mutations cause disease.

3. Generic Name of Product: Long Read Sequencing kits

Salient characteristics All deliverables required under this contract shall be packaged, marked and shipped in accordance with Government specifications. At a minimum, all deliverables shall be marked with the contract number and Contractor name. The Contractor shall guarantee that all required materials shall be delivered in immediate usable and acceptable condition.

Vendor must be able to provide these specifications:

a. Delivers paired full-length T-cell receptor alpha and beta chain sequences at the single cell level using universal and unbiased amplification strategies, thus avoiding amplification and detection bias associated with complex multiplex PCR while revealing deeper biological complexity of antigen specificity on a per-cell basis than single-chain (e.g. TCRβ) methods.

Version 11-30-2016-1 FAR Part 11—Describing Agency Needs

b. The incorporation of a unique molecular identifier (UMI) in the template switch oligo enables transcript quantification and further reduces systematic bias by controlling for non-biological SNVs or other variants introduced by PCR and sequencing error.

c. Enables the detection of germline and somatic variation across full-length antigen receptor V(D)J gene sequences.

d. Identifies both productive and non-productive recombination events as well adaptive immune cells with more than one productive T-cell receptor alpha or beta chain.

e. Maps transcriptions start sites.

5. Delivery Date: One (1) month after receipt of completed purchase order

6. Period of Performance: Not applicable

7. Option(s): Not applicable
