Version 11-30-2016-1 FAR Part 11—Describing Agency Needs

PURCHASE DESCRIPTION

1. Statement of Need and Purpose: The National Institute on Aging (NIA) has a Zeiss 780 confocal microscope that is nearing obsolescence, and as such, NIA requires a replacement with a new confocal microscope. This replacement is critical in maintaining productivity in scientific research within the Laboratory of Neurogenetics, and amongst internal/external collaborators.

2. Background Information and Objective: This replacement will maintain The National Institute on Aging’s productivity by combining excellent multiplex imaging performance with low phototoxicity and high-speed scanning. Upgraded microscopy software will allow regions of interest to be found quickly and consistently, while direct processing of images will enable parallel acquisition and rapid data processing. Most importantly, a new instrument will assure that workflow will not be impeded if the current system requires extensive repairs or ceases to function entirely.

3. Generic Name of Product: Inverted confocal microscope

4. Salient characteristics All deliverables required under this contract shall be packaged, marked and shipped in accordance with government specifications. At a minimum, all deliverables shall be marked with the contract number and contractor name. The contractor shall guarantee that all required materials shall be delivered in immediate usable and acceptable condition. Vendor must be able to provide the following specifications for the Zeiss LSM 980 with Airyscan and 8 detector scan head (or equal):

• LSM 980 8 detector scanhead must provide, at the minimum:

o Switching Mirror R3.5 o Scan module RGB f/LSM 980 o Spectral Detector 2PMT+4CH GaAsP o Secondary beam splitter wheel for DC-out at LSM 980 o Module power supply, electronics and laser (488,561,639nm) o External detector NIR f/LSM

• Inverted stand with Definite Focus 3 must provide:

o Axio Observer 7 SP stand for LSM 980 with DF3 nosepiece o Axio Observer 7 with motorized focus drive (min. step width 10 nm), path deflection to the tube, motorized 3-position optovar turret, 6-position nosepiece o Reflected, fluorescence illumination manual Set for LSM o Scanning Stage 130x100 STEP with stepper motor (2 mm spindle pitch)

• AI sample finder and camera must provide:

o AI Sample Finder - with motor operated aperture diaphragm and 7 turret disc positions for H, Ph1, Ph2, Ph3, 2x DIC as well as additional position for autofocus and coherence contrast Axio Observer 7 with motorized focus drive (min. step width 10 nm), path deflection to the tube, motorized 3-position optovar turret, 6-position nosepiece o High performance microscopy monochrome camera incl. driver software 64bit, Interface card USB 3.1 Gen2 PCIe x4, dual USB 3.0 cable 3 m o Camera Operation: Exposure time adjustment with continuous, automatic or manual mode, Adjustable over or under-exposure mode, Optional measurement on defined target area (spot-meter), Image sharpening, Dark current compensation, Single pixel noise correction.

o Supported Operating Systems: for ZEN: Windows 10 x64 Enterprise Supported Application Software: ZEN 3.6 (blue edition) or higher, ZEN 3.5 core or higher The required application software is provided via ZEISS Microscopy Installer or ZEISS Download Center.

• Fast Piezo Z must provide:

o Z-Piezo stage insert WSB 500 (D) including control electronics, controller and cable. Z range 500 micrometer.

o Stage insert Z PIEZO WSB 500 for Heating Inserts P S1 / Mxx S1 (D) to insert into Stage attachment Z PIEZO WSB 500 on inverted microscope stands. Fixing to stage attachment through magnets.

• Objectives must provide:

o Objective Plan-Apochromat 20x/0.8 M27 (FWD=0.55mm) o Objective Plan-Apochomat 40x/1.3 Oil DIC M27 (WD=0.20mm), UV-IR incl.

Immersol 518 F for 30°

• Airtable must be:

o System table, mid size, air damped, level regulated pressured air supply necessary, long side open , with breadboard, 1200x900mm

• Airyscan SuperResolution detector must provide:

o Patented superresolution method based on 32 concentrically arranged GaAsP detection elements. The design enables acquisition with superresolution as well as confocal resolution. Airyscan 2 provides a 4- 8 times higher sensitivity compared to single GaAsP detectors. In contrast to confocal imaging a 2 times higher resolution in lateral an axial direction can be achieved with the SR mode.

The higher resolution and sensitivity can be combined with a markedly increased imaging speed without losing image quality.

• Accessories must include:

o ZEISS Predictive Service – Remote condition monitoring program to improve instrument uptime

• Imaging computer must provide:

o Microscopy Workstation Premium (HP Z8 G5) (O) HP Z8 Fury G5 Workstation -

Chipset: Intel W790 - Memory Expansion Slots: 16 x DDR5, RDIMM, ECC, 4800 MHz Modules - Memory: max. 1024 GB RAM

• Imaging and peocessing software must in clude the following:

o ZEN 3.9 Image acquisition and processing under Windows 10 x64 user interface configurable, control of the ZEISS microscope systems and components, extensive acquisition and analysis, CZI image format. The following functions must be included: - arivis cloud advanced - Basic 3D viewing - colocalization -connect basic - direct processing - manual EDF - measurement - panorama -spectral unmixing - ZEN Data Storage Client - acquisition base (time series and multichannel).

5. Quantity: One complete system as detailed above

6. Delivery Date: Delivery date of 6-12 months after receipt of completed purchase order

7. Period of Performance: Manufacturer warranty for one calendar year beginning date of receipt
