Solicitationno:NIEHS2013158November2012

STATEMENT OF WORK

IndependentlyandnotasanagentoftheGovernment,theContractorshallfurnish
services,qualifiedpersonnel,material,equipment,andfacilities,nototherwise
providedbythegovernmentunderthetermsofthiscontractasneededtoperform
theworksetforthbelow.
Thiscontractwillprovidesupporttothegovernmenttoexamineavariablenumber
ofchemicalsannually,dependingontheneedsofthegovernmentandavailable
funding,forinductionofgeneticchanges(e.g.,mutations,chromosomalchanges,
DNAdamage)inanimalorhumancells.Approximately25invivoassaysmaybe
conductedannually.Thecontractwillalsoprovidethegovernmentthecapabilityto
examineupto5chemicalsforabilitytoinducegeneticchanges(e.g.,chromosomal
changes,DNAdamage)inculturedmammaliancells.Inadditiontoeukaryotictest
systems,thecontractwillprovidethecapabilitytotestapproximately15chemicals
peryearforinductionofgenemutationsinbacterialtesterstrains.TheContractor
musthavethecapabilityofconductingtheassaysdescribedinthisStatementof
Work(SOW)onsite.Thenumberandtypeofassaysmayvaryfromyeartoyear,
andthecontractormusthavetheabilitytoconductmultipleassaysatthesame

time.

1.Chemicalstestedforpotentialgenotoxicityunderthiscontractwillbesupplied
bytheCORthroughtheNTPchemistrycontractsorwillbeidentifiedbytheCORfor
purchasebytheContractor.
2.TheContractorshallperformallworkinaccordancewithstandardoperating
procedures(SOPs)andtestarticlespecificprotocolsapprovedbytheCOR.
Standardprotocolsforeachassaytypemustbeinplacebeforetestingisinitiated,
andshallbeapprovedbytheCORwithin30daysaftertheawardismade.Foreach
chemical,theCORwillidentifythespecificassaystobeperformed.Notall
chemicalsortestarticleswillbetestedineveryassay.
3.TheContractorshallhaveawrittenQualityManagementPlanavailablefor
inspectionbytheCOR(see:http://www.epa.gov/quality/qmps.html).The
ContractorshallkeepalllaboratoryrecordsinaccordancewithFDAGood
LaboratoryPracticeRegulations(originallypublishedinFederalRegister,1978,pp.
(21CFRSection58)).TheContractorshallperformallworkinamanner
compatiblewiththeattachedNTPHealthandSafetyMinimumRequirements.
4.TheCORwilldeterminethetestarticlestobeexaminedandtheassay(s)tobe
used.Testprotocolsandprotocolamendmentswillbeapprovedbysignatureofthe

COR.TheCORmaychangetheprotocolstokeepcurrentwithstateoftheart
technologyorimprovements/changesinassaydesign,ortomeetspecificProgram
needs.Assaytypesshallincludebutarenotlimitedto:
Assay1–invivomicronucleusassay,exposuresconductedatan
alternatelaboratoryorsite
TheContractorshallmeasuremicronucleatederythrocyte,reticulocyte,
and/orlymphocytefrequenciesincodedbonemarroworbloodsamples
obtainedfromanimals,typicallymiceandrats,oroccasionallyhumans
exposedtoachemical(s)inanalternatelaboratoryorsite.Typically,the
animalexposureswillbeconductedatanNTPcontractlaboratorythat
conductsshort‐termtoxicity,2‐yearcancerbioassay,orotherstudiesin
rodents.Bloodsamplesfromtheseanimalswillbecollected,chilled,and
packagedforovernightshipmentoncoldpackstotheContractor.If
necessary,toexpeditereceipt,theContractormaychoosetocollectthe
packagedsamplesatthecourier’sfacilityandtransportthemtotheContract
Laboratorytoensuresamplequality.UponarrivalattheContract
Laboratory,thesampleswillbeprocessedandstoredinaminus80°freezer.
Typically,sampleswillbeevaluatedusingflowcytometricprocedures.A
noncommercialcollaboratinglaboratory,withinoroutsidetheNIH(e.g.,NCI,
CDC,DOD)typicallyprovidescodedhumanbloodsamplestobeevaluatedfor
micronuclei.Nopersonalidentifiersshallbeattachedtoanyhumansamples.
Datafromallmicronucleusstudiesshallbeelectronicallyentereddirectly
intoacontrolled‐accessNTP‐hosteddatabaseforevaluationand
interpretationbytheCOR.TheNTPshallprovideaccesstothecomputer
systemandtrainingintheuseofthecomputersystem.

Assay 2 – in vivo micronucleus assay, exposures conducted by the Contractor

TheContractorshallmeasuremicronucleatederythrocyte,reticulocyte,
and/orlymphocytefrequenciesinblood(oroccasionallybonemarrow)
samplesobtainedfromanimals,typicallymiceandrats,exposedtoa
chemical(s)attheContractLaboratory.Treatmentregimensandsample
timeswillbedeterminedbytheCOR.Onoccasion,asdirectedbytheCOR,
otherdividingcellpopulationsmaybeassessedforfrequencyofmicronuclei.
ProceduresshallbeconsistentwiththosedescribedinOECDGuideline474.
Samplesshallbecollected,processed,storedasnecessary,andevaluated.For
thesein‐housestudies,theContractorshallperformdose‐rangingstudies
unlesstheCORinstructsotherwise.TheContractorshallperformallassays
usingappropriatesolventandpositivecontrols(whereappropriate).
Flexibilityinmethodsofdatacollection(slidesorflowcytometry)isa
requirement,althoughemphasiswillbeontheuseofflowcytometryfor
evaluatingfrequencyofmicronucleatedcells.Datahandlingwillbeas

describedforAssay1above,withoneaddition.Tocapturethein‐lifeportion
ofthedatafromthistypeofassay,theContractorshalluseanNTP‐hosted
computersystemforwhichaccessandtrainingwillbeprovidedbyNTP.

Assay 3 – in vivo comet assay

TheContractorshallquantitateDNAdamage(e.g.,singleanddoublestrand
breaks,oxidativedamage,alkalilabilesites,andDNA‐DNAandDNA‐protein
crosslinks)usingthesinglecellgelelectrophoresis(cometassay)methodin
tissuesdesignatedbytheCORinanimals,typicallymiceandrats,or
occasionallyhumansexposedtoachemical(s).Nopersonalidentifiersshall
beattachedtoanyhumansamples.Exposuresmayoccureitherinan
alternatelaboratoryorsite(e.g.,anNTPcontractlaboratory),orfor
laboratoryanimalsonly,intheContractLaboratory.Treatmentregimensand
samplecollectiontimesmayvary.Examplesoftissuesorcellsthatmaybe
examinedforchemical‐inducedDNAdamageinanimalsincludebutarenot
limitedtobloodleukocytes,liver,kidney,stomach,ileum,colon,andbrain.
Humansampleswouldbelimitedtoperipheralblood.Singlecell
suspensionsofsamplesgeneratedinanalternatelaboratoryshallbe
preparedatthatlaboratoryandfrozenatminus80°Cinabalancedsalt
solutionsuppliedbytheContractor.Subsequently,thecodedsamplesshall
beshippedbyovernightcouriertotheContractor;theContractorshall
collectthematthecourier’sfacility,transportthemtotheContract
laboratory,cataloguethesamples,andstoretheminaminus80°Cfreezer
untiltheyarethawedandevaluated.Tissuesamplesobtainedfromanimals
exposedattheContractLaboratoryshallbepreparedappropriately
(methodsdifferbytissuetype)andeitheranalyzedimmediatelyorfrozenat
minus80°Cforfutureanalysis.Slidesshallbepreparedandelectrophoresed
followingpublishedmethods.Animageanalysissystemshallbeusedto
locateandevaluatecellsforDNAdamage.Electronicimagesofeachcellthat
isevaluatedshallbestored.Alldata,includingcellimages,shallbedeposited
electronicallyintoanNTPdatabase(seeAssay1,above).Cometdatashall
alsobecapturedbytheimageanalysissystemusedbytheContractorand
transferredtoexcelspreadsheets.Thereisanadditionalrequirementthat
theContractorperformacytotoxicitymeasurementfortissuesselectedfor
DNAdamageassessment.Tocapturethein‐lifeportionofthedatafrom
studiesinwhichexposureswereconductedattheContractLaboratory,the
ContractorshalluseanNTP‐hostedcomputersystemforwhichaccessand
trainingwillbeprovidedbytheNTP.

Assay 4 – combined in vivo micronucleus/comet assay

TheContractorshallsimultaneouslymeasuremicronucleusfrequenciesin
erythrocytesand/orreticulocytes,andDNAdamageintissuesselectedby
theCOR,inlaboratoryanimals(typicallymiceorrats)exposedtoa

chemical(s)attheContractLaboratory.Thiscombinedmicronucleus/comet
assayshalladheretopublishedprotocolsalthoughalternate
dosing/samplingregimensmaybespecifiedbytheCORforparticular
chemicals.BloodandtissuesamplesshallbecollectedintheContract
Laboratoryandprocessedasappropriatefortheendpoint.Bloodsamples
(and/orbonemarrowsamples)formicronucleusanalysisshallbeprepared
asdescribedforAssays1and2;tissuesamplesforthecometassayshallbe
preparedandanalyzedasdescribedforAssay3.Datafromthemicronucleus
andcometstudiesshallbeelectronicallyentereddirectlyintothecontrolled‐
accessNTP‐hostedgenetictoxicitydatabaseforevaluationand
interpretationbytheCOR.Tocapturethein‐lifeportionofthestudydata,the
ContractorshalluseanNTP‐hostedcomputersystemforwhichaccessand
trainingwillbeprovidedbytheNTP.

Assay 5 – Pig‐a gene mutation assay

TheContractorshallmeasurethefrequencyofX‐linkedPig‐a
(phosphatidylinositolglycan,ClassA ) genemutationsinperipheralblood
erythrocytesandreticulocytesobtainedfromanimals,typicallymiceand
rats,oroccasionallyhumans,exposedtoachemical(s).Animalexposures
mayoccurattheContractor’slaboratoryoranalternatelaboratorysuchas
anNTPcontractlaboratory.Anoncommercialcollaboratinglaboratory,
withinoroutsidetheNIH(e.g.,NCI,CDC,DOD)typicallyprovidescoded
humanbloodsamplestobeevaluatedforfrequencyofcellsbearingPig‐a
mutations.Nopersonalidentifiersshallbeattachedtoanyhumansamples.
Fortheanimalstudies,peripheralbloodsamplesshallbeobtainedfromeach
animalusinganapprovedmethodandbloodsamplesshallbeimmediately
processedfollowingacceptedprotocolsforevaluationofthefrequencyof
cellswithaberrantGPIanchorexpression(CD59‐negativecells),whichisa
directmeasureofPig‐agenemutation.DataonthefrequencyofPig‐a
mutantcells(typicallyreticulocytesanderythrocytes)shallbesubmitted
electronicallyinawrittenreporttotheCOR.

Assay 6 – in vitro micronucleus assay

TheContractorshallmeasurethefrequencyofmicronucleatedcellsin
culture,followingtreatmentwithatestarticleinthepresenceandthe
absenceofexogenousmetabolicactivation.Theprocedurefollowedbythe
ContractorshallbeconsistentwiththatoutlinedinOECDguideline487.The
testarticleandcelltypewillbechosenbytheCOR.Aninitialrangefinding
studyshallbeconductedtosettestarticleconcentrations.Cytotoxicitymust
bequantifiedintherangefindingstudyandtoxicitylevelsshallnotbe
excessiveatthehighestconcentrationselectedforthedefinitivestudy.In
thedefinitivestudy,appropriatevehicleandpositivecontrolsshallbe
included.TheuseofcytochalasinBshallbeconsidered,particularlyfor

asynchronouslydividingcellpopulations.Datashallbesubmitted
electronicallyinawrittenreporttotheCOR.

Assay 7 – in vitro comet assay

TheContractorshallmeasurechemical‐inducedDNAdamageincultured
cellsusingthesinglecellgelelectrophoresis(cometassay)method.Cells
shallbetreatedandexaminedintheContractLaboratory.Celltypeandtest
articlewillbedeterminedbytheCOR.Thedurationofchemicalexposureas
wellasothertechnicalaspectsoftheexposureandslidepreparations,
includingtheconcentrationoflowdensityagaroseandtheelectrophoresis
characteristics,shallbeselectedbytheCORinconsultationwiththe
Contractor.Insomecases,theCORmayrequestthataspecificprotocol
designedtomeasurechemical‐inducedoxidativedamagebefollowed.Cells
shallbeevaluatedforDNAdamageusinganimageanalysisplatform.Data
shallbesubmittedelectronicallytotheNTPcometassaydatabase.

Assay 8 – bacterial mutagenicity assay

TheContractorshallmeasuregenemutationfrequencies(evaluatedas
numberofmutantcoloniesperplate)intesterstrainsofSalmonella
typhimuriumand/orEscherichiacolifollowingchemicalexposurescarried
outwithandwithoutasourceofexogenousmetabolicactivation.Thetest
article(s)shallbeprovidedbytheNTP.Concentrationrangefindingstudies
shallbeconductedtoselecttestconcentrations.Appropriatevehicleand
positivecontrolsshallbeincludedineachexperiment.Examinationof
chemicalsinS.typhimuriumstrainsTAl00,TA98,andtheE.colistrainWP2
(uvrA,pKM101)isacorerequirementforeachbacterialmutagenicityassay.
TestsusingadditionalordifferentstrainsmayberequestedbytheCORto
meetspecificinformationalneedsoftheGovernment.TheContractorshall
performatleastonerepeatexperimenttoconfirmtheresultsfromtheinitial
experiment.TheCORshalldeterminewhichexperimentsdonotmeet
qualitycontrolcriteriaandwhennecessary,shallrequestadditionalrepeat
testsbeyondthetwotrialsspecified.Datashallbesubmittedelectronicallyto
theNTPbacterialmutagenicitydatabase.

Assay development and protocol refinement

Inadditiontotheassaysdescribedabove,asdirectedbytheCOR,the
Contractorshallengageinresearchanddevelopmentactivitiesrequiredfor
assaydevelopmentandrefinementaspertinenttechnologyimprovements
andinnovationsoccurduringthecourseofthiscontract.Forexample,over
thepastdecade,methodsformeasuringmicronucleatederythrocyte
frequencieschangedfromprimarilyslide‐baseddataacquisitiontouseof
flowcytometryforevaluatingbloodsamples.Thischangeindataacquisition

methodsnecessitateddevelopmentofexpertiseinflowcytometricanalysis
ofbloodsamplesanddemonstrationthatresultsfromflowcytometric
analyseswereconsistentwithresultsfromslide‐basedmethodsinbothrats
andmice.Thesetypesofresearchanddevelopmentactivitiesmayrequire
upto550hrofthetotaleffortonanannualbasis.
5.ThePrincipalInvestigatorandotherkey/criticalpersonnelshallvisittheNIEHS
toconsultwiththeCORandotherNIEHSstaff,asdirectedbytheCOR.Inaddition,
thePrincipalInvestigatorshallattendonescientificmeetingorconferenceannually
topresentdataanddiscussresultsofgenetictoxicitytestsperformedunderthe
contract,asdirectedbytheCOR.Onoccasion,keyorcriticalpersonnel,asdefined
inthisSOW,maybeaskedbytheCORtotraveltoacquirespecificcontract‐related
trainingtoenhancetechnologicalcapabilitiesasmethodsusedinthefieldadvance.
Training‐relatedtravelrequirementsmayvaryfromyeartoyear,butarenot
anticipatedtoexceedonetripperyearforonememberoftheContractLaboratory
staff.Arequiredtrainingsessionshouldbeconsideredasasingleday,runningfrom
8AMto5PM.Inaddition,theCORmaydirectthatonememberoftheContractor’s
stafftraveltoanotherNTPcontractlaboratoryorothersiteforoneormore
workdaysforthepurposeofprovidingtrainingortocollecttissuesamplesfor
analysisbytheContractor.
6.AsdirectedbytheCOR,theContractorshallshipanimaltissues,slides,orother
biologicalmaterialorinformationincluding(butnotlimitedto)protocolsanddata
totheNIEHSortoanotherlocationintheU.S.Someoftheshipmentsmayrequire
dryiceorshipmentonweticeorcoldpacks.
7.TheGovernmentshallarrangeforreceiptoftissuesorcellsamplesassociated
withconductingtheassaysdescribedinthisSOW.TheGovernmentshallalso
providetheB6C3F1micerequiredforanyoftheinvivostudiesdescribedinthis
SOW;HarlanSpragueDawleyratsshallbepurchasedbytheContractorasneeded
fortheinvivoassaysdescribedinthisSOW.Onoccasion,theContractormaybe
directedbytheCORtoobtainanimalsfromoutsidesourcestobeusedinassays
conductedattheContractLaboratory.
8.Celllinesandbacterialstrainstobeusedintheinvitrogenetictoxicitytestsshall
beobtainedbytheContractor,atthedirectionoftheCOR.
9.Allfacilities,practices,andproceduresofhusbandry,care,andmaintenanceof
animalsusedintheperformanceofthiscontractshallbeinaccordancewiththe
latestissueineffectofthe''GuidefortheCareandUseofLaboratoryAnimals."The
mostcurrentversion,theEighthEdition,(ReferenceHHSAR370.401and

10.TheContractorshallbeaccreditedbytheAssociationfortheAssessmentand
AccreditationofLaboratoryAnimalCare(AAALAC)International.TheContractor’s
animalcareandusecommittee(ACUC)shallapprovetheanimalcareanduse
protocolsforallassaysconductedwithanimalsinperformanceofthiscontract.The
ACUC‐approvedprotocolsshallbesubmittedtotheCORpriortotheinitiationof

studies.

DELIVERABLES/TECHNICAL REQUIREMENTS

TheContractorshallenterresultsfromassaysforwhichanNTPelectronicdata
capturesystemisavailable,directlyintothedatabase.Theresultsfromtheassays
shallbeenteredwithin30daysofthecompletionoftesting.TheContractor
shallnotrecorddataonpapertobeenteredatalaterdateunlesstheNTPserveris
unavailable.TheContractorshallnotincludethesedataintheannualorfinal
reports(seebelow).Toaccomplishdataentry,theContractormusthave:
I.AMacintoshcomputeroraPCrunningthemostrecentoperatingsystemsand
webbrowsers(SafariorExplorer)available,withcapabilitytoupgradeasnewer
softwareprogramsbecomeavailable.

2. An Internet connection: high‐speed cable access is required.

3.OtherSoftware:AdobeAcrobatReaderandMicrosoftExcel(aspartofthe
MicrosoftOfficesuiteofsoftware)inthemostadvancedversionsavailable.

4. An NTP hosted computer system to capture the in-life portion of the study data. The NTP shall provide access to the computer system and training in the use of the computer system.

Note:theinternetbrowsersandAcrobatreaderarefreesoftware.Therequired
softwareshouldbeupgradedassoftwareimprovementsbecomeavailable.The
aboveequipmentandsoftwareisrequiredateachworkstationwherea
technicianisrecordingdatasothatthedatacanbeentereddirectlyintotheweb
application.Thisequipment,software,andinternetaccessibilityisalsorequired
fortheLaboratoryDirector,thePrincipalInvestigator,andallkeypersonnel.

REPORTING REQUIREMENTS AND DELIVERABLES

ReportingofNTPdataasindicatedbythestatementofwork:
TheContractorshallenterresultsfromassaysforwhichanNTPelectronicdata
capturesystemisavailabledirectlyintotheNTPdatabase.Theresultsfromthe
assaysshallbeenteredwithin30daysofthecompletionoftesting.TheContractor
shallnotrecorddatageneratedfromtheseassaysonpapertobeenteredatalater
dateunlesstheNTPserverconnectionisunavailable.TheContractorshallnot
includethesedataintheannualorfinalreports(describedbelow).

1. Studyreports
ForeachassayconductedwithatestarticlefortheNTPattheContract
Laboratory,awrittenreportshallbesubmitted.Thisreportshallsummarize
theprotocolthatwasfollowed;includinganyapprovedamendments,and
shallsummarizetheresultsandotherimportantfindingsandobservations
(dataanalysisisnotrequired,asallstudydatawillbeanalyzedbytheNTP).
ForthoseassaysthatdonothaveanNTPelectronicdatacapturesystem
available,therawdataandthesummarydatashallbeincludedinthestudy
report.Withthisreport,asummaryofcostsassociatedwitheachstudyshall
besubmitted.Thesecostsshallinclude,butnotbelimitedtoatotalperassay
cost,aswellasmaterials,supplies,shippingcosts,etc.associatedwitheach
individualstudy.

2. Biannual/Annual Reports

a) TheContractorshallsubmitbiannual(6‐month)reportsbeginning6
monthsafterthedateofcontractawardandattheendofeach12month
periodofperformance.
b) BiannualandAnnualreportsshallincludeasummaryofresultsfrom
eachchemical/assayconducted,statusofchemicalsontestortobe
tested,problemsencountered,problemresolution,andanticipated
problems.TheBiannualandAnnualreportsshallnotcontaintheassay

data.

3. Final Reports

TheContractorshallelectronicallysubmitafinalreportwithin30daysof
completionofthecontract.TheFinalreportshallincludeasummaryofall
samplestestedandtheassayresults.

4. Summary of Salient Results

TheContractorisrequiredtoprepareandsubmit,withthefinalreport,a
summary(nottoexceed200words)ofsalientresultsachievedduringthe
performanceofthecontract.Thisreportshallbedueonorbeforethe
expirationdateofthecontract.
TheContractorshallsubmit6‐month,annualandfinalreportselectronically,
onecopytotheCORandonetotheContractingOfficial,asaPDFunless
otherwisedirectedbytheCOR.AnelectroniccopyoftheDraftFinalReport
shallbeduetotheCOTR60daysbeforethecompletiondateofthecontract,
withtheFinalReportdueatthetimeofcontractcompletion.
Allelectronicallysubmittedreportsmustbe508compliant.

Personnel needs to be met by the contractor: See RFP Section L

Oversightofanimalexperiments:
ContractormusthaveanexperiencedAnimalCareandUseCommittee(ACUC)for
approvalofinvivoanimalexperimentsinadditiontoshowingstrictadherenceto
NIHguidelinesforanimalcareanduse.Thecontractormustalsohaveanapproved
animalwelfareassuranceplanonfilewiththeNIHOfficeofAnimalWelfarepriorto
contractaward.TheContractormustbeincompliancewithPublicHealthService
policiesconcerninganimal‐relatedactivities.Forallin‐lifeanimalstudiesconducted
attheContractLaboratoryasdescribedinthisSOW,euthanasiashallbe
accomplishedby70:30CO2/O2anesthesiafollowedbyexsanguinationtoensure
deathpriortosamplecollection.

Invoicing schedule
