FAR Part 11—Describing Agency Needs Revised, November 11, 2016

PURCHASE DESCRIPTION

1. Statement of Need and Purpose:

Procurement of a Confocal Laser Scanning Microscope System for high quality fluorescent optical slice imaging and 3D reconstruction of mouse brain tissues.

2. Background Information and Objective:

Hattar Lab studies circadian rythms using mouse brains. Histological processing, fluorescent immunohistochemical staining, fluorescent optical slice imaging and

3D reconstruction of mouse brain tissues are routine jobs for the lab. The majority of our data collection work relies on a high quality Confocal Laser

Scanning Microscope. The purpose of the component is to accomplish fluorescent brain tissue imaging with high fluorophore sensitivity, large field of view, high resolution, fast tiling speed and versatile 3D reconstruction.

5. Functional / Performance Specifications

1. Super high sensitivity for fluorescence detection utilizing two basic technologies: a) hexagonal continuously variable pinhole, which achieves equivalent confocality to a square pinhole at 1.2 Airy units, optimizing confocality with minimal light loss. b) low-angle incidence method utilized on the dichroic mirrors which increases fluorescence efficiency by 30%.

2. Super high imaging resolution of up to 4096 x 4096 pixels.

3. Super accurate real time spectral unmixing during a fast imaging process.

Government requires a maximum performance in the separation of closely overlapping fluorescence spectra and the elimination of autofluorescence during the acquisition of a 32-channel spectral image (512 x 512 pixels) with a single scan in 0.6 second. Moreover, We also need 512 x 32-pixel images captured at 24 fps while maintaining the super accurate real time spectral unmixing.

4. Spectral imaging across wide band, achieved by simultaneous excitation with four lasers selected from a maximum of eight wavelengths.

5. High-sensitivity spectral image delivered by a GaAsP PMT, the A1-DUVB tunable emission detector.

6. 25mm field of view

7. Fast tiling speed of 10 fps (at 512 x 512 pixels) and 24 fps (at 512 x 32-pixels).

A good resolution needs to be maintained by scanner driving and sampling systems and image correction technology.

8. User-friendly software system to be able to integrate the Ti2-E inverted microscope system, TIRF system, spectral detector and Perfect Focus System. All systems must be controlled by a single platform.
