SOLICITATION NUMBER:
SPECIFICATIONS
Surface Plasmon Resonance (SPR) Bio-molecular Binding Kinetics Characterization System
The Naval Research Laboratory (NRL) requires a label-free interaction analysis system based on the principle of surface plasmon resonance for biomolecular binding measurements. The system must be multiplexed, allowing for the parallel injection of solutions over a single sensor chip, and provide both kinetic and affinity binding information over the specified range of association and dissociation kinetic rate constants. In addition to these general requirements, the following specifications are required:
1. The system must accommodate both 96- and 384-well plate providing 1500 uL or more of volume for injection. This is required for our current high throughput work in testing antibody binding affinity for a wide range of antibodies, ligands and conditions.
2. The system must have a temperature controlled sample storage area with temperature range from 4 oC to 39 oC (or higher) and an independently controlled analysis temperature range which also ranges from 4 oC to 39 oC (or higher). This is required for our current heat shock related investigation studies and controls.
3. The system must be capable of measuring association rate constants 5x108 M-1s-1 or higher for our proteins of interest and measuring dissociation rate of 1x10-5 s-1 or lower.
4. For our fast dissociation studies we require the system be capable of measuring dissociation rate constant 0.4 s-1 or higher
5. Based on our current investigation and plan to study low molecular weight binding species we require the baseline noise be typically less than 0.03 RU (RMS) where RU stands for refractive index unit.
6. The measurable refractive index must range from 1.33 to 1.38 (or higher)
7. The system must employ at least 6 or more injection needles allowing for 6 or more injections of both ligand and analyte over a single chip.
8. The injection needle design must have two fluidic channels per needle allowing for both capture and reference data to be collected from the same needle.
9. The system must have a two-solution cycle option in which two different solutions are introduced over the surface in a single cycle allowing for high-throughput buffer composition studies.
10. The system must be able to operate for more than 48 hours unattended for high throughput studies of protein/ligand interactions
11. The manufacturer must offer compatible bare gold sensor chips for user customizable surface studies.
12. The system must include the option of introducing analyte in one cycle (no regeneration of surface needed) or in multiple cycles in which the surface is chemically regenerated to make sure the measurements are not incorporating artifacts from the regeneration process.
13. System must accommodate a wide range of sample types including proteins, RNA, DNA in media ranging from simple buffers to complex media such as plasma and serum.
14. The injection volume must be as low as 1 uL for investigations of expensive or difficult to purify proteins. In order to rule out diffusive artifacts in the data, flow rates of up to 100 uL/min (or greater) are required.
15. The system must include software for aiding in experimental design as well as data analysis.
DELIVERY: The Contractor shall deliver to NRL, all components making up a complete Surface Plasmon Resonance (SPR) Bio-molecular Binding Kinetics Characterization System no later than ninety (90) days after contract award.
INSTALLATION: The Contractor shall perform system installation within three (3) weeks after receipt of the complete Surface Plasmon Resonance (SPR) Bio-molecular Binding Kinetics Characterization System by NRL.
TRAINING: The Contractor shall provide 1-2 days of on-site training within two weeks of system installation. Training will be provided to at least four (4) Government employees.
WARRANTY: The Contractor shall provide a one (1) year on-site system warranty from the date of installation by a company authorized installer.
