PART 3 – TECHNICAL REQUIREMENTS

3.1. BACKGROUND

Due to a significant increase in the number on foodborne outbreaks associated with consumptions of a variety of fresh produce, the Division of Microbiology in the Office of

Regulatory Science has devoted the past 10 years to the study of the contamination, attachment, and internalization of human enteric pathogens into fresh produce. Laser Scanning Confocal

Microscopy is the key instrument to study the mechanisms of pathogen attachment and persistence on and inside of fresh fruits and vegetable. The detection and analysis of the internalization of bacterial and protozoan pathogens into fresh produce commodities is impossible with any other equipment because vegetal tissues have extremely high level and broad spectrum of autofluorescence that overlaps with fluorescence spectra of virtually all currently used fluorescent antibodies and fluorescent proteins used to tag bacteria. Zeiss LSM

880, 34-Channel System with Airyscan allows the linear unmixing, extreme sensitivity and superresolution that will allow us to overcome the limitations of working with vegetal matrices and permit unprecedented detection, identification, and characterization of human enteric pathogen internalized into food matrices. Furthermore, human enteric pathogens are capable of persisting in food processing plants by colonizing plastic, glass or stainless steel surfaces via biofilm formation. It is crucially important to study this biofilm formation using the Confocal

Laser Scanning Microscopic approach.

Various surfaces will be incubated in appropriate media, with or without food residues, for up to

7 days to promote biofilm formation. Planktonic cells will be rinsed off using a sodium chloride solution. Biofilm cells will be stained with appropriate fluorescent dyes and the confocal microscope will be used to distinguish between live and dead cells. The biofilm structure will also be disrupted and the released cells will be used to infect an epithelial human cell monolayer.

The confocal microscope will be used to determine the compartmentalization of bacteria within the cell monolayers. This will allow us to determine what effect biofilm formation had on the ability of pathogens to maintain virulence capabilities.

The understanding of the mechanism of bacteria and protozoa adherence and colonization host cells, foods and food- contact surfaces is critical for the FDA mission in preventing foodborne illnesses.

LASER SCANNING CONFOCAL 34 CHANNEL SYSTEM FEATURES:

• A 34-Channel Quasar Detection unit, consisting of a calibrated 32-element Gallium Arsenide Phosphide (GaAsP) array Photomultiplier Tube with typical QE >45 % (peak) and two calibrated flanking single Photomultiplier Tube detectors; the one covering the long wavelength range has to be peltier cooled. Has to enable image acquisition with freely definable emission windows and advanced spectral acquisition and analysis. The detector array shall enable three distinct modes of spectral image acquisition;

• A flexible multi-channel configuration utilizing the combination of prisms and plates allowing the user to define a collection window per detector with 1 nm resolution

• Allow simultaneous 34 detector spectral acquisition with 9.7nm spectral resolution (two side flanking Photomultiplier Tubes in addition to the 32-element Photomultiplier Tube array).

• Provide sequential high resolution spectral scanning that utilizes user selectable fixed width detection slits (2.9 or 4.6 nm widths) transverse the emission spectrum to build up the emission spectrum. (This will ensure that the user collects repeatable and accurate emission spectrum).

• A microlens array situated in front of the 32-element linear Gallium Arsenide Phosphide array Photomultiplier Tube to ensure there is no cross talk between detector channels and provide accurate spectral information.

• An actively cooled the 32-element linear Gallium Arsenide Phosphide (GaAsP) array Photomultiplier Tube (PMT) and actively cooled red enhanced Photomultiplier Tube to allow Fluorescence-Cross-Correlation-Spectroscopy (FCCS) supporting up to 6 simultaneous channels.

• Digital oversampling imaging mode that allows for the user to control a high voltage gain and digital offset on the detection Photomultiplier Tube.

• Photon counting imaging mode that allows for the system to distinguish the arrival of single photons many tens of times during pixel dwell time as an image is scanned (GaAsP PMTs and cooled PMTs are run at 15MHz). (This mode shall permit the user to discriminate actual photon arrivals events to the detector and allows for samples with low amounts of fluorocromes to be easily imaged and produce a noise free image).

• TwinGate main beamsplitter utilizing 2 separate 13 position MBS wheels allowing up to 50 different laser combinations while maintaining a typical laser suppression of Optical Density 7 or greater.

• Highly efficient holographic grating leads to an even spectral dispersion over the entire visual spectral range (380-750 nm). Grating shall utilize a ‘spectral recycling loop’ to recover non-separated light, redirecting it onto the holographic grating for optimum sensitivity.

• Pigtailed Lasers, that are upgradeable, requires no maintenance and is stable.

• Software integrated with linear modulation of laser power from 0.001 to 100% for all laser lines between 458 and 633nm. (This is high critical for the user as this will allow to reach the low light level imaging powers that are required for sensitive experiments).

The Contractor shall comply with the provisions of this paragraph (d) if this contract was awarded using other than sealed bid, is in excess of the simplified acquisition threshold, and does not contain the clause at 52.215-2, Audit and Records-Negotiation.

(1) The Comptroller General of the United States, or an authorized representative of the Comptroller General, shall have access to and right to examine any of the Contractor’s directly pertinent records involving transactions related to this contract.

(2) The Contractor shall make available at its offices at all reasonable times the records, materials, and other evidence for examination, audit, or reproduction, until 3 years after final payment under this contract or for any shorter period specified in FAR Subpart 4.7, Contractor Records Retention, of the other clauses of this contract. Records relating to appeals under the disputes clause or to litigation or the settlement of claims arising under or relating to this contract shall be made available until such appeals, litigation, or claims are finally resolved.

(e)(1) Notwithstanding the requirements of the clauses in paragraphs (a), (b), (c), and (d) of this clause, the Contractor is not required to flow down any FAR clause, other than those in this paragraph (e)(1) in a subcontract for commercial items. (iii) 52.222-17, Nondisplacement of Qualified Workers (May 2014) (E.O. 13495). Flow down required in accordance with paragraph (l) of FAR clause 52.222-17.
