Part 3 – Description/Specifications BRAND NAME or EQUIVALENT IncuCyte® S3 Live-Cell Analysis System

3.1 Background

The development of induced pluripotent stem cells and of novel engineered in vitro systems, such as organoids and microphysiological systems, have high potential to predict clinical effects of drugs in vitro. However, cellular systems present high levels of variability at the different following levels: system-to-system, lab-to-lab and batch-to-batch. To deal with phenotypical variability, understand it and deal with it in our experiments, we need to monitor the development and changes in cellular systems. This system will allow the different researchers in the Division to monitor stem cells, microphysiological systems and other cellular systems and to gain information on their development and biological properties as a function of time in culture.

3.2 Statement of Work/Salient Characteristics

The proposed instrument must be newly manufactured unit, not used or refurbished, or previously used for demonstration.

This equipment consists of an imaging system installed inside of a cell culture incubator and allows imaging and tracking cells in culture in real time cell for long periods of time (up to months). This equipment is connected to a computer and allows for tracking many plates and many transparent cell culture devices simultaneously. This equipment also allows saving, processing and deriving cellular phenotypes from these acquired videos. The IncuCyte automatically acquires and analyzes images in real time, providing an information-rich analysis on the cells being cultured. It has capability to image cells in brightfield or to image cells labeled with fluorescent markers. Our Division is strongly investing its research efforts into new cellular-based approaches to predict the clinical outcomes of drugs.

Salient Characteristics:

· Be a microscope that operates inside of a cell culture incubator.

· The system must contain a camera to acquire images of cells.

· Be able to image cells on different types of transparent cell culture platforms: Petri dishes, flasks, well plates, etc.

· The system must acquire red and green fluorescent channels.

· Have at least 5x, 10x and 20x magnifications.

· The system must be connected to a computer to save and process acquired images.

· Can be remotely controlled.

· The system must image cells in culture.

· The system must be able to image cells different cell culture platforms and in different positions within the same culture platform.

Extra Components:

· Computer (Windows 10), the cables that connect the computer to the system and software

· Microscope must have a camera to acquire images of samples.

· Stage must be automatic in x, y and z Pacing adapter/cables must be provided

· Must have phase contrast

· Must be user friendly

· Must be designed to avoid contaminations and maintain a sterile environment.

· Must have fluorescence capabilities.

Delivery, Installation, Training, and Support: The vendor shall cover all costs associated with the following:

· Crating and shipping shall be included in this proposal to the delivery address.

· On-site installation, hardware, software, and applications training shall be provided at the FDA site.

· Installation and a minimum of 1 day on-site training must be included.

· All appropriate training manuals and literature must be supplied either electronically or in hard copy.

· Technical support shall be provided via common avenues: phone, web and/or fax during standard business hours.

Warranty/Preventative Maintenance Service Agreement Following initial warranty/service/technical support coverage, the FDA shall have a unilateral right to exercise optional periods to extend the warranty/service agreement to include preventative maintenance. Option to purchase instrument service contract from instrument manufacturer is required because use of alternative service provider will void the warranty of these instruments.

· The instrument manufacturer shall provide an extended service plan to include annual preventative maintenance and service contracts for up to 2 years after the initial instrument warranty period.
