Part 3 – Description/Specifications/Work Statement

3.1 Scope of Work

The purpose of this contract is to purchase (1) Automated Patch Clamp System.

Electrophysiology is the study of the electrical properties of cells and tissues in the the body. It involves recording voltage signals or current flow resulting from ions moving across ion channel proteins in the cell membrane. Because functions of excitable cells in the heart and the brain rely on the cells’ abilities to generate precise electrical signals, drugs and diseases that affect ion channels in these tissues will alter cardiac and brain functions.

The Division of Applied Regulatory Science at the Food and Drug Administration is building an “Ion Channel Shared Resource Facility” to generate the necessary electrophysiology data for assessing adverse effects of drugs and diseases on cells in the heart and the brain. An essential equipment of this facility is an automated patch clamp system that can 1) record electrophysiology signals from different isolated cell types (e.g. cell lines, stem cells, and dissociated cardiac myocytes/neurons), and 2) produce high quality data comparable to those generated using manual patch clamp systems by highly skilled electrophysiologists. To enable data acquisition even by non-expert users, an automated patch clamp is needed.

3.2 Specifications:

The vendor shall provide an automated patch clamp system, including the amplifier and data acquisition/analysis software. The software shall operate under the 64-bit Windows 7 operating system. The automated patch clamp system shall meet all required criteria listed below. It shall be able to:

a) Acquire single cell measurements (no population patch recording).

b) Acquire data from at least 4 cells simultaneously.

c) Perform current-clamp and voltage-clamp recordings from different types of isolated cells, including cultured cell lines, transiently transfected cultured cells, and induced pluripotent stem cell-derived cells such as cardiac myocytes.

d) Consistently achieve seal resistance no less than 1 GΩ without using seal enhancers. Success with >1 GΩ seal when using cultured cell lines shall be greater than 70%.

e) Variable speed for temperature ramps, allowing the recording temperature to be increased at a user-defined speed from room temperature to ~37ºC during the middle of an experiment. This feature – the ability to change temperature during the middle of an experiment – is needed to ensure data compatibility with existing manual patch clamp systems in the Division of Applied Regulatory Science.

f) Perform automated series resistance and whole cell capacitance compensation at the on-set of experiment. Additionally, this system shall allow the user to increase the degree of compensation during the middle of an experiment if series resistance were to change, or the system shall be able to check series resistance at various time points during the experiment and automatically adjust the degree of compensation accordingly.

g) Shall have continuous perfusion capability (microfluidic format only, no open-well format), allowing different concentrations of drugs to be applied; shall be able to perform fast drug application (<10 ms), allowing drug wash-in and wash-out kinetics to be measured.

h) Easily programmable, allowing user to define and adjust experimental protocols.

i) Modular – can be upgraded easily to increase data throughput.

j) Amplifier(s) shall be included.

-One Year Warranty shall be included.

-Customer service with onsite and online training and technical support.

- Assistance with experimental protocol troubleshooting or system replacement if data acquired consistently fall short of the manufacturer-stated system capabilities.
